摘要:
摘 要 目的:探讨丹酚酸B对代谢记忆环境下离体培养的RSC96细胞凋亡及炎性因子变化规律的影响。方法:RSC96细胞分别培养于低糖培养组(56 mmol/L)、代谢记忆(125 mmol/L高糖培养48 h,56 mmol/L低糖培养24 h)、丹酚酸B组(125 mmol/L高糖+1 μmol/L丹酚酸B培养48 h,56 mmol/L低糖培养基+1 μmol/L丹酚酸B培养24h)中,流式法检测ROS含量,Western blot法检测Cleavedcaspase 9,Cleavedcaspase 3,P65,IL1β的表达。结果:代谢记忆环境可下调ROS含量,下调Cleavedcaspase 9,Cleavedcaspase 3,P65,IL1β等蛋白的表达;丹酚酸B可上调ROS含量,下调Cleavedcaspase 9,Cleavedcaspase 3,P65,IL1β等蛋白的表达。结论:代谢记忆环境下,丹酚酸B可能通过下调ROS引起细胞凋亡及炎性损伤保护RSC96细胞。
Abstract:
Abstract Objective:To observe the effect of Sal B on apoptosis and inflammatory of RSC96 cultured in metabolic memory environmentMethods:RSC96 cells were respectively cultured in control group(56mmol/L),metabolic memory(125mmol/L high glucose for 48h plus 56mmol/L glucose for 24 h) or Sal B group(125mmol/L high glucose + 1μmol/L Sal B for 48h plus 56mmol/L glucose+ 1μmol/L Sal B for 24 h)Flow cytometry was performed to detect ROS levelWestern blot was manipulated to detect the expression of Cleavedcaspase 9,Cleavedcaspase 3,P65,IL1βResults:The metabolic memory group ROS level was upregulated compared with the control groupHowever,Sal B group ROS level was downregulated compared with the metabolic memory groupThe expression of Cleavedcaspase 9,Cleavedcaspase 3,P65,IL1β in metabolic memory group decreased compared with the control groupHowever,the expression of Cleavedcaspase 9,Cleavedcaspase 3,P65,IL1β in Sal B decreased compared with the metabolic memory groupConclusion:Sal B protected RSC96 cells against apoptosis and inflammatory by eliminating ROS in metabolic memory environment
