摘要:
目的:探讨非小细胞肺癌(NSCLC)组织中长链非编码RNA(lncRNA)DiGeorge综合征关键区域基因5(DGCR5)、乳腺癌易感基因1(BRCA1)、T细胞免疫球蛋白及黏蛋白结构域分子-1(TIM-1)表达与患者临床病理特征和预后的相关性。方法:选取NSCLC患者120例,术中收集患者癌组织及癌旁组织标本,比较组织中lncRNA DGCR5、BRCA1、TIM-1表达,分析癌组织中三者表达与患者临床病理特征的关系。随访3年,分析lncRNA DGCR5及BRCA1、TIM-1蛋白表达与患者3年生存率的关系。多因素Cox回归分析NSCLC患者预后的影响因素。结果:与癌旁组织比较,NSCLC组织中TIM-1蛋白阳性表达率升高,lncRNA DGCR5表达水平、BRCA1蛋白阳性表达率降低(均P<0.05)。NSCLC组织中lncRNA DGCR5、BRCA1、TIM-1表达与TNM分期、淋巴结转移、肿瘤分化程度有关(均P<0.05)。lncRNA DGCR5低表达患者3年生存率低于高表达患者,BRCA1阴性患者3年生存率低于BRCA1阳性患者,TIM-1阳性患者3年生存率低于TIM-1阴性患者(均P<0.05)。NSCLC患者3年内生存的危险因素为肿瘤低分化、淋巴结转移、TNM分期Ⅲ期、lncRNA DGCR5低表达、BRCA1蛋白阴性表达、TIM-1蛋白阳性表达(均P<0.05)。结论:NSCLC组织中lncRNA DGCR5、BRCA1、TIM-1异常表达,且三者与NSCLC患者临床病理特征及预后有关。
Abstract:
Objective:To investigate the correlation between the expression of long non-coding RNA DiGeorge syndrome critical region gene 5 (lncRNA DGCR5),breast cancer susceptibility gene 1 (BRCA1),and T cell immunoglobulin and mucin-domain containing molecule-1 (TIM-1) in non-small cell lung cancer (NSCLC) tissue and the clinicopathological characteristics and prognosis of patients.Methods:A total of 120 patients with NSCLC were enrolled.Cancer tissue and adjacent non-cancerous tissue specimens were collected during surgery,and the expression levels of lncRNA DGCR5,BRCA1 and TIM-1 were compared.The relationships between the expression of these three factors in cancerous tissues and the clinicopathological characteristics of patients were analyzed.Patients were followed up for 3 years,and the relationship between the expression of lncRNA DGCR5,BRCA1,and TIM-1 and the 3-year survival rate was analyzed.Multivariate Cox regression was used to identify independent prognostic factors.Results:Compared with adjacent non-cancerous tissues,NSCLC tissues showed increased positive expression rate of TIM-1 protein and decreased expression levels of lncRNA DGCR5 and positive expression rate of BRCA1 protein (all P<0.05).The expression of lncRNA DGCR5,BRCA1 and TIM-1 in NSCLC tissues was significantly associated with TNM stage,lymph node metastasis,and tumor differentiation degree (all P<0.05).Patients with low expression of lncRNA DGCR5 had a lower 3-year survival rate than those with high expression;patients with negative BRCA1 expression had a lower survival rate than those with positive expression;and patients with positive TIM-1 expression had a lower survival rate than those with negative expression (all P<0.05).Poor tumor differentiation,lymph node metastasis,TNM stage Ⅲ,low expression of lncRNA DGCR5,negative BRCA1 protein expression,and positive TIM-1 protein expression were identified as risk factors for 3-year survival in NSCLC patients (all P < 0.05).Conclusion:The abnormal expression of lncRNA DGCR5,BRCA1 and TIM-1 in NSCLC tissues is closely related to the clinicopathological characteristics and prognosis of NSCLC patients.
