摘要:
目的:探讨微小RNA-21(miR-21)对宫颈癌细胞恶性生物学行为的影响及可能的作用机制。方法:将宫颈癌HeLa细胞随机分为对照组(空白未处理)、miR-NC组(转染miR-NC)、miR-21 mimics组(转染miR-21 mimics)、miR-21 inhibitor组(转染miR-21 inhibitor)。采用流式细胞术、CCK-8法、Transwell实验、细胞划痕实验分别检测HeLa细胞凋亡、增殖、侵袭、迁移情况;RT-qPCR检测HeLa细胞中miR-21表达;Western blot检测HeLa细胞中磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)/哺乳动物雷帕霉素靶蛋白(mTOR)信号通路相关蛋白表达。结果:与对照组和miR-NC组比较,miR-21 mimics组HeLa细胞凋亡率下降,细胞增殖率、侵袭数量及迁移率升高;miR-21 inhibitor组HeLa细胞凋亡率升高,细胞增殖率、侵袭数量及迁移率降低(均P<0.05)。对照组和miR-NC组HeLa细胞中miR-21及p-PI3K、p-AKT、p-mTOR蛋白表达水平低于miR-21 mimics组,高于miR-21 inhibitor组(均P<0.05)。结论:抑制miR-21能够促进宫颈癌细胞凋亡,降低细胞增殖、侵袭及迁移能力,其机制可能与调控PI3K/AKT/mTOR信号通路有关。
Abstract:
Objective:To investigate the effects of microRNA-21 (miR-21) on the malignant biological behavior of cervical cancer cells and its potential mechanism of action.Methods:Cervical cancer HeLa cells were randomly divided into four groups:the control group (untreated),the miR-NC group (transfected with miR-NC),the miR-21 mimics group (transfected with miR-21 mimics),and the miR-21 inhibitor group (transfected with miR-21 inhibitor).Flow cytometry,CCK-8 assay,Transwell assay and cell scratch assay were respectively used to detect apoptosis,proliferation,invasion and migration of HeLa cells.RT-qPCR was used to measure the expression of miR-21 in HeLa cells,while Western blot was used to detect the expression of proteins related to the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (AKT)/mammalian target of rapamycin (mTOR) signaling pathway.Results:Compared with the control and miR-NC groups,the miR-21 mimics group exhibited decreased apoptosis rates and increased proliferation rates,invasive numbers,and migration rates of HeLa cells (all P<0.05).Conversely,the miR-21 inhibitor group showed increased apoptosis rates and decreased proliferation rates,invasive numbers,and migration rates (all P<0.05).The expression levels of miR-21 and p-PI3K,p-AKT,p-mTOR proteins in HeLa cells of the control group and miR-NC group were lower than those in the miR-21 mimics group and higher than those in the miR-21 inhibitor group (all P<0.05).Conclusion:Inhibiting miR-21 can promote the apoptosis of cervical cancer cells,reduce the proliferation,invasion and migration abilities of the cells,and the mechanism may be related to the regulation of the PI3K/AKT/mTOR signaling pathway.
