摘要:
目的:探讨长链非编码RNA(lncRNA)LINC01370对黑色素瘤细胞增殖和侵袭的影响及其作用机制。方法:采用Cancer LncRNA Census数据库比较黑色素瘤组织和正常皮肤组织中LINC01370表达差异。RT-qPCR检测人表皮黑色素细胞HEM和黑色素瘤细胞(VMM5A、WM266-4、A2058、A-375)中LINC01370的表达水平,选取表达水平最低的细胞进行后续实验。将LINC01370高表达质粒和对照质粒转染至A2058细胞,分为LINC01370组和NC组。采用克隆形成实验检测A2058细胞增殖能力,Transwell实验检测A2058细胞能力,Western blot检测A2058细胞中增殖和侵袭蛋白表达。双荧光素酶报告基因实验验证LINC01370和miR-1246的靶向关系。采用RT-qPCR检测A2058细胞中miR-1246表达。结果:LINC01370在黑色素瘤组织中的表达水平低于正常皮肤组织(P<0.01)。LINC01370在VMM5A、WM266-4、A2058、A-375细胞中的表达水平低于HEM细胞,且A2058细胞表达水平最低(均P<0.05)。与NC组比较,LINC01370组A2058细胞LINC01370表达水平升高(P<0.01)。与NC组比较,LINC01370组A2058细胞增殖能力、侵袭细胞数以及细胞周期蛋白依赖性激酶3(Cdk3)、细胞周期蛋白C(Cyclin C)、转化生长因子-β(TGF-β)、叉头框C2(FOXC2)蛋白表达水平降低(均P<0.01)。miR-1246与LINC01370-WT质粒共转染的相对荧光素酶活性低于miR-NC与LINC01370-WT质粒共转染(P<0.01)。与NC组比较,LINC01370组A2058细胞miR-1246表达水平降低(P<0.01)。结论:LINC01370在黑色素瘤组织及细胞中低表达,LINC01370可能通过敲低miR-1246表达抑制黑色素瘤细胞的增殖和侵袭。
Abstract:
Objective:To investigate the effect of lncRNA LINC01370 on the proliferation and invasion of melanoma cells and its mechanisms.Methods:The Cancer LncRNA Census database was utilized to compare the expression differences of LINC01370 between melanoma tissues and normal skin tissues.RT-qPCR was employed to detect the expression levels of LINC01370 in human epidermal melanocytes(HEM)and melanoma cells(VMM5A,WM266-4,A2058,A-375),with the cells showing the lowest expression levels selected for subsequent experiments.LINC01370 overexpression plasmids and control plasmids were transfected into A2058 cells,which were then divided into LINC01370 and NC groups.Colony formation assays were conducted to assess the proliferative capacity of A2058 cells,Transwell assays to evaluate the invasive capabilities,and Western blot to determine the expression of proteins associated with proliferation and invasion in A2058 cells.Dual-luciferase reporter gene assays were used to verify the targeting relationship between LINC01370 and miR-1246.RT-qPCR was also used to detect the expression of miR-1246 in A2058 cells.Results:The expression level of LINC01370 in melanoma tissues was lower than that in normal skin tissues(P<0.01).The expression level of LINC01370 in VMM5A,WM266-4,A2058 and A-375 cells was lower than that in HEM cells,and the expression level in A2058 cells was the lowest(all P<0.01).Compared with NC group,the expression level of LINC01370 in A2058 cells was increased in LINC01370 group(P<0.05).Compared with the NC group,the proliferation ability of A2058 cells,the number of invasive cells and the expression levels of cyclin-dependent kinase 3(Cdk3),Cyclin C,transforming growth factor-β(TGF-β)and forkhead box C2(FOXC2)proteins in the LINC01370 group were decreased(all P<0.01).The relative luciferase activity of miR-1246 co-transfected with LINC01370-WT plasmid was lower than that of miR-NC co-transfected with LINC01370-WT plasmid(P<0.01).Compared with NC group,the expression level of miR-1246 in A2058 cells in LINC01370 group was decreased(P<0.01).Conclusion:LINC01370 is underexpressed in melanoma tissues and cells,and it may inhibit the proliferation and invasion of melanoma cells by downregulating the expression of miR-1246.
