摘要:
目的:探讨早产新生儿胎盘和脐带组织血管紧张素Ⅱ 1型受体(AT1R)和血管紧张素Ⅱ 2型受体(AT2R)表达及临床意义。方法:选取早产儿32例为早产儿组,另选取同期出生的健康足月儿30例为足月儿组,比较两组新生儿一般资料。收集胎盘组织、脐带组织和脐血,酶联免疫吸附法(ELISA)检测脐血肾素-血管紧张素系统(RAS)关键成分,实时荧光定量PCR和免疫组织化学法检测AT1R、AT2R mRNA和蛋白表达。结果:足月儿组胎龄、体重、身长、头围、胎盘重量、体重指数、1 min 和5 min Apgar评分高于早产儿组(均P<0.05)。足月儿组脐血血管紧张素转化酶2(ACE2)和血管紧张素1-7(Ang 1-7)水平高于早产儿组(均P<0.05)。早产儿组胎盘组织AT1R、AT2R mRNA表达水平高于足月儿组(均P<0.05)。早产儿组胎盘和脐带组织AT1R、AT2R蛋白表达高于足月儿组(均P<0.05)。结论:AT1R、AT2R在早产儿胎盘、脐带组织中高表达,AT1R、AT2R可能影响胎组织器官的发育和成熟,与早产密切相关。
Abstract:
Objective:To investigate the expression and clinical significance of angiotensinⅡ1 receptor(AT1R)and angiotensin Ⅱ 2 receptor(AT2R)in placenta and umbilical cord tissue of premature infants.Methods:Thirty-two preterm infants were selected as the preterm infants group,and 30 healthy full-term infants born at the same time were selected as the full-term infants group,and the general data were compared.The placenta tissue,umbilical cord tissue and umbilical cord blood of the two groups were collected.The key components of RAS in umbilical cord blood were detected by ELISA.The mRNA and protein expressions of AT1R and AT2R were detected by real-time PCR and immunohistochemistry.Results:Gestational age,body weight,body length,head circumference,placental weight,BMI,1 and 5minutes Apgar scores in the full-term infants group were higher than those in the preterm infants group(all P<0.05).The levels of ACE2 and Ang 1-7 in umbilical cord blood in full-term infants group were higher than those in preterm infants group(all P<0.05).Compared with full-term infants group,the mRNA expressions of AT1R and AT2R in the placenta tissue of preterm infants group were higher(all P<0.05).Compared with full-term infants group,AT1R and AT2R protein expressions in placenta and umbilical cord tissue of preterm infants group were higher(all P<0.05).Conclusion:AT1R and AT2R are highly expressed in the placenta and umbilical cord tissue of preterm infants.AT1R and AT2R may affect the development and maturation of fetal tissues and organs,and are closely related to preterm birth.
