摘要:
目的:建立脂质代谢相关食管鳞癌(ESCC)预后预测模型,并通过体外细胞实验进行验证。方法:利用差异分析和Lasso-Cox回归分析筛选ESCC脂质代谢相关基因并构建预后预测模型,利用Kaplan-Meier曲线、单多因素Cox回归分析、受试者工作特征(ROC)曲线及临床病例特征分析风险评分的独立预后预测能力,通过体外实验验证PTPDC1在ESCC细胞中的表达和功能。结果:共筛选出4个与ESCC患者预后显著相关的脂代谢基因,可作为分子标签将ESCC患者分为高、低风险组。Kaplan-Meier曲线分析结果显示,低风险组患者总生存期(OS)显著优于高风险组(P<0.001)。单多因素Cox回归分析显示,由4个脂质代谢相关基因构成的风险评分可作为ESCC患者预后预测的独立因素。ROC曲线分析显示,脂质代谢风险评分的预后预测效能最佳。实时荧光定量PCR(RT-qPCR)结果显示,与人正常食管HET-1a细胞比较,PTPDC1在ESCC细胞系ECA109和KYSE150中的表达水平显著升高(均P<0.05)。敲降PTPDC1后,si-1-PTPDC1组和si-2-PTPDC1组PTPDC1相对表达量明显低于NC组(均P<0.05); 与NC组比较,ECA109和KYSE150细胞增殖和迁移能力显著降低(均P<0.05)。结论:基于4个脂质代谢基因构建的预后预测模型具有较好的预后预测价值,可以辅助ESCC患者进行个体化治疗。
Abstract:
Objective:To develop a prognostic prediction model for lipid metabolism-associated esophageal squamous carcinoma(ESCC),and verify it by in vitro cell experiments.Methods:Difference-in-difference and Lassos-Cox regression analysis were used to screen genes related to lipid metabolism of ESCC and construct a prognostic prediction model.Kaplan-Meier curve,univariate and multivariate Cox regression analysis,ROC curve and clinical case characteristics were used to analyze the independent prognostic prediction ability of risk score.The expression and function of PTPDC1 in ESCC cells were verified by in vitro experiments.Results:The screening obtained four lipid metabolism genes were significantly associated with the prognostic prediction of ESCC patients,which could be used as molecular labels to categorize ESCC patients into high- and low-risk groups.The results of the Kaplan-Meier curve analysis showed that patients in the low-risk group had a significantly better overall survival(OS)than that in the high-risk group(P<0.001).Univariate and multivariate Cox regression analysis showed that the risk score consisting of four lipid metabolism-related genes could be used as an independent factor for prognostic prediction of ESCC patients.ROC curves analysis showed that the lipid metabolism risk score had the best prognostic predictive efficacy.RT-qPCR results showed that the expression levels of PTPDC1 in ESCC cell lines ECA109 and KYSE150 were significantly higher than those in human normal esophageal HET-1a cells(both P<0.05).After PTPDC1 knockdown,the relative expression of PTPDC1 in si-1-PTPDC1 group and si-2-PTPDC1 group was significantly lower than that in NC group(all P<0.05); compared with NC group,the proliferation and migration ability of ECA109 and KYSE150 cells were significantly decreased(all P<0.05).Conclusion:The prognostic prediction model based on the four lipid metabolism genes has good prognostic value and can assist the individualized treatment of ESCC patients.
