摘要:
目的:探讨血管内皮生长因子受体3(VEGFR-3)、基质金属蛋白酶-9(MMP-9)、nm23-H1蛋白在口腔癌组织中的表达及其临床价值。方法:选取行手术治疗的口腔鳞状细胞癌患者70例,收集癌组织标本。另选取同期体检健康者70例,采集口腔黏膜组织。采用免疫组织化学染色法检测样本组织VEGFR-3、MMP-9、nm23-H1蛋白表达,分析其与患者临床病理特征的相关性。术后随访5年,记录患者生存情况。分析VEGFR-3、MMP-9、nm23-H1蛋白表达与患者术后5年预后的关系。结果:口腔癌组织VEGFR-3、MMP-9蛋白阳性表达率高于口腔黏膜组织,nm23-H1蛋白阳性表达率低于口腔黏膜组织(均P<0.05)。VEGFR-3、MMP-9蛋白阳性表达患者淋巴结转移比例较高(均P<0.05)。nm23-H1蛋白阳性表达患者淋巴结转移、TNM分期Ⅲ期-Ⅳ期及肿瘤浸润深度>5 mm比例较低(均P<0.05)。口腔癌组织VEGFR-3、MMP-9蛋白表达与淋巴结转移呈正相关(均P<0.05)。m23-H1蛋白表达与淋巴结转移、TNM分期、肿瘤浸润深度呈负相关(均P<0.05)。VEGFR-3、MMP-9蛋白阳性表达患者5年生存率低于阴性表达患者(均P<0.05)。m23-H1蛋白阳性表达患者5年生存率高于阴性表达患者(P<0.05)。结论:口腔癌组织VEGFR-3、MMP-9蛋白高表达,nm23-H1蛋白低表达,三者与淋巴结转移以及口腔癌患者预后有关。
Abstract:
Objective:To explore the expression of vascular endothelial growth factor receptor 3(VEGFR-3),matrix metalloproteinase-9(MMP-9)and nm23-H1 protein in oral cancer tissue and their clinical significance.Methods:Seventy patients with oral squamous cell carcinoma who underwent surgical treatment were selected,and cancer tissue specimens were collected.Additionally,oral mucosal tissues were collected from 70 healthy individuals during the same period.Immunohistochemical staining was used to detect the expression of VEGFR-3,MMP-9 and nm23-H1 proteins in the tissue samples,and their correlation with the clinical pathological characteristics of the patients was analyzed.The patients were followed up for 5 years postoperatively to record their survival status and to analyze the relationship between the expression of VEGFR-3,MMP-9 and nm23-H1 proteins and the patients' prognosis after 5 years.Results:The positive expression rates of VEGFR-3 and MMP-9 proteins in oral cancer tissue were higher than those in oral mucosal tissue,while the positive expression rate of nm23-H1 protein was lower than that in oral mucosal tissue(all P<0.05).Patients with positive expression of VEGFR-3 and MMP-9 proteins had a higher proportion of lymph node metastasis(all P<0.05).Patients with positive expression of nm23-H1 protein had a lower proportion of lymph node metastasis,TNM stage Ⅲ-Ⅳ and tumor invasion depth >5 mm(all P<0.05).The expression of VEGFR-3 and MMP-9 proteins in oral cancer tissue was positively correlated with lymph node metastasis(all P<0.05).The expression of nm23-H1 protein was negatively correlated with lymph node metastasis,TNM stage,and tumor invasion depth(all P<0.05).Patients with positive expression of VEGFR-3 and MMP-9 proteins had a lower 5-year survival rate than those with negative expression(all P<0.05).Patients with positive expression of nm23-H1 protein had a higher 5-year survival rate than those with negative expression(P<0.05).Conclusion:High expression of VEGFR-3 and MMP-9 proteins and low expression of nm23-H1 protein in oral cancer tissue are associated with lymph node metastasis and the prognosis of patients with oral cancer.
