摘要:
目的:探讨血必净调控核因子E2相关因子2(Nrf2)/抗氧化反应元件(ARE)信号通路对百草枯中毒大鼠肺组织纤维化的保护作用。方法:选择雄性SD大鼠30只,分为对照组、模型组、血必净组,每组各10只。建立百草枯中毒大鼠模型,血必净组腹腔注射血必净注射液4 ml/kg,1 d/1次,连续3 d,对照组和模型组大鼠腹腔注射等体积生理盐水。苏木精-伊红(HE)染色、Masson染色观察肺组织结构变化,并进行组织病理评分,肺组织中氧化应激指标及胶原蛋白含量,实时荧光定量PCR(qRT-PCR)和蛋白质免疫印迹(Western blot)对肺组织核因子相关因子-2(Nrf2),抗氧化反应元件(ARE)mRNA和蛋白表达进行测定。结果:肺组织结构方面,对照组大鼠无异常,且无肺纤维化改变和炎症存在,而模型组大鼠肺泡结构异常,并伴有明显炎性变化以及大量增殖的肺间质纤维细胞,胶原蛋白沉积增多。与模型组比较,血必净组大鼠组肺组织炎性改变和胶原蛋白沉积减少。对照组、模型组、血必净组肺组织病理评分依次为0、(5.52±0.49)、(2.31±0.34)分,血必净组肺组织病理评分低于模型组(t=17.020,均P<0.001)。与对照组比较,模型组肺组织丙二醛(MDA)、羟脯氨酸(HYP)升高,超氧化物歧化酶(SOD)均下降,与模型组比较,血必净组肺组织SOD升高,MDA、HYP下降(均P<0.05)。与对照组比较,模型组肺组织Nrf2、血红素氧含酶1(HO-1)mRNA和蛋白表达均升高,血必净组肺组织Nrf2、HO-1 mRNA和蛋白表达高于模型组(均P<0.05)。结论:血必净注射液可能通过上调Nrf2-ARE信号通路,减轻百草枯诱导的大鼠肺纤维化程度。
Abstract:
Objective:To investigate the protective effect of Xuebijing on pulmonary fibrosis in rats with paraquat poisoning by regulating Nrf2-ARE signaling pathway.Methods:Thirty male SD rats were selected and divided into control group,model group and Xuebijing group with 10 rats each.The rat model of paraquat poisoning was established.The rats in the Xuebijing group were intraperitoneally injected with Xuebijing injection(4 ml/kg,1 time/1 day)for 3 consecutive days,and the rats in the control group and the model group were intraperitoneally injected with the same volume of normal saline.Hematoxyn-eosin(HE)and Masson staining were used to observe the structural changes of lung tissue,and histopathological score,oxidative stress index and collagen content in lung tissue were performed.Real-time quantitative fluorescent PCR(qRT-PCR)and Western blot were used to determine the mRNA and protein expression of nuclear factor-associated factor-2(Nrf2)and heme oxygenase-1(HO-1)in lung tissue.Results:The lung tissue structure of rats in the control group was normal,without inflammation and fibrosis changes,while the pulmonary alveolar cavity of rats in the model group had obvious inflammatory changes,local alveolar structure was abnormal,lung interstitial fibrocyte proliferation and collagen deposition occurred.Compared with the model group,the changes of lung inflammation and collagen deposition were reduced in Xuebijing group.The pathological scores of lung tissue in control group,model group and Xuebijing group were 0 points,5.52±0.49 points,2.31±0.34 points,and the pathological scores of lung tissue in Xuebijing group were lower than those in model group(t=17.020,P<0.001).Compared with the control group,MDA and HYP were increased and SOD were decreased in lung tissue of model group.Compared with the model group,SOD was increased and MDA and HYP were decreased in lung tissue of Xuebijing group(all P<0.05).Compared with the control group,the mRNA and protein expressions of Nrf2 and HO-1 in lung tissues of model group were increased,and those of Xuebijing group were higher than those of model group(all P<0.05).Conclusion:Xuebijing injection may reduce the degree of paraquat-induced pulmonary fibrosis in rats by up-regulating Nrf2-ARE signaling pathway.
