摘要:
目的:探究促红细胞生成素对正畸牙复发大鼠牙周组织改建及Wnt/β-连环蛋白(β-catenin)信号通路的影响。方法:构建大鼠正畸牙移动模型,将48只正畸牙模型大鼠随机分为模型组、促红细胞生成素低(250 IU/kg)、高(500 IU/kg)剂量组、促红细胞生成素(500 IU/kg)+KYA1797K(Wnt/β-catenin抑制剂,25 mg/kg)组,每组12只; 分组后拆除各组大鼠加力装置,另取12只健康大鼠作为对照组(不进行任何处理)。各组大鼠给予相应干预30 d。计算取下加力装置第10、20、30天大鼠正畸牙复发率; 苏木素-伊红染色观察牙周组织病理学变化; 免疫组织化学染色检测牙周组织骨形态发生蛋白(BMP)2、BMP4表达; 荧光定量PCR法和蛋白印迹法分别检测牙周组织Wnt、β-catenin mRNA及蛋白表达水平。结果:对照组大鼠牙周组织中纤维排列紧密,细胞整齐分布; 与对照组相比,模型组牙周组织中纤维分散,细胞间排列间隙大,牙槽骨表面有少量成骨细胞排列,没有明显的细胞牙骨质,各时间段正畸牙复发率、牙周组织BMP2、BMP4表达、Wnt、β-catenin mRNA和蛋白水平显著升高(均P<0.05); 与模型组相比,促红细胞生成素低、高剂量组大鼠牙周组织中纤维分布逐渐密集,成骨细胞和细胞牙骨质逐渐增多,各时间段正畸牙复发率依次降低,牙周组织BMP2、BMP4表达、Wnt、β-catenin mRNA和蛋白水平依次升高(均P<0.05); KYA1797K能部分逆转高剂量促红细胞生成素对大鼠正畸牙复发的改善作用(均P<0.05)。结论:促红细胞生成素可能通过激活Wnt/β-catenin信号通路,加速骨质生成,促进牙周组织改建,减缓大鼠正畸牙复发。
Abstract:
Objective:To investigate the effects of erythropoietin on periodontal tissue remodeling and Wnt/β-catenin signaling pathway in rats with orthodontic tooth recurrence.Methods:A rat orthodontic tooth movement model was established,48 orthodontic tooth model rats were randomly divided into model group,erythropoietin low- dose group(250 IU/kg),erythropoietin high-dose group(500 IU/kg),and erythropoietin(500 IU/kg)+KYA1797K(Wnt/β-catenin inhibitor,25 mg/kg)group,12 rats in each group.After grouping,the force device was removed in each group,and another 12 healthy rats were taken as control group(without any treatment).Each group of rats was given corresponding intervention for 30 days.The orthodontic tooth recurrence rate was calculated at 10,20,and 30 days after removing the force device.Hematoxylin-eosin staining was used to observe the pathological changes of periodontal tissue.Immunohistochemical staining was used to detect the expression of bone morphogenetic protein(BMP)2 and BMP4 in the periodontal tissue.The levels of Wnt,β-catenin mRNA and protein in periodontal tissues were detected by fluorescence quantitative PCR and Western blot.Results:The fibers in the periodontal tissue of control group rats are tightly arranged and the cells were evenly distributed.Compared with control group,the fibers in the periodontal tissue of model group were dispersed,the intercellular arrangement space was larger,a few osteoblasts were arranged on the alveolar bone surface,and there was no obvious cellular cementum,the orthodontic tooth recurrence rate in different time periods,the expressions of BMP2 and BMP4,the levels of Wnt,β-catenin mRNA and protein in periodontal tissue significantly increased(all P<0.05).Compared with model group,the fiber distribution in periodontal tissue of rats in erythropoietin low- and high-dose groups was gradually dense,and osteoblasts and cellular cementum were gradually increased,the orthodontic tooth recurrence rate in different time periods were decreased in turn,and the expressions of BMP2,BMP4,the levels of Wnt,β-catenin mRNA and protein in periodontal tissue were increased in turn(all P<0.05).KYA1797K could partially reverse the improvement effect of high dose erythropoietin on orthodontic tooth recurrence in rats(P<0.05).Conclusion:Erythropoietin may accelerate bone formation,promote periodontal tissue remodeling and slow orthodontic tooth recurrence in rats by activating Wnt/β-catenin signaling pathway.
