摘要:
目的:探讨通过整合DNA修复能力和核苷酸剪切修复基因表达来评估头颈部鳞状细胞癌(HNSCC)的发病风险。方法:82例HNSCC患者作为研究对象,选择性别和年龄与患者匹配的81例健康人作为正常对照组,研究对象均来自中国汉族人群。使用宿主细胞再活化(HCR)实验来测定HNSCC患者和对照者的DNA修复能力(DRC); 使用qPCR实验检测HNSCC患者和对照组外周血淋巴细胞中9个核心NER基因的mRNA相对表达水平; 使用Logistic回归分析DRC和NER mRNA表达水平之间的相关性; 使用受试者工作特征曲线(ROC)评估整合DRC和NER mRNA表达水平对HNSCC风险预测的效能。结果:HNSCC患者DRC的平均水平(9.84±2.32)%明显低于对照组(10.35±2.42)%(P=0.013)。DRC水平与NER基因mRNA表达水平相关性研究发现DRC与XPA之间存在关联(P=0.031)。与单一水平相比,DRC水平曲线下面积显著提高(P=0.043)。同时,将NER mRNA表达水平整合到ROC曲线中,与仅含DRC的水平相比,含DRC和XPA mRNA的水平曲线下面积明显提高(P=0.046)。结论:整合DRC和NER mRNA表达对HNSCC发病风险的预测效能显著提高。相比于只包含一种表型的水平,整合两种标志物能更有效地评估HNSCC的发病风险。
