摘要:
目的:探究栀子苷(GE)在部分膀胱出口梗阻(pBOO)模型大鼠中的治疗作用及机制。方法:将大鼠随机分为假手术(Sham)组,pBOO组,GE 20、40、80 mg/kg组,每组6只。pBOO手术后4周,连续灌胃给药GE,1次/d,共2周。实验终点,检测GE对pBOO模型大鼠膀胱肌张力和膀胱尿流动力学的影响,利用HE染色评价GE对pBOO大鼠膀胱组织炎性反应的改善作用,检测GE对pBOO大鼠膀胱组织丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)的影响,蛋白印迹和实时荧光定量PCR检测GE对Nrf2/NF-κB通路蛋白表达的影响。结果:GE 40、80 mg/kg组可以显著降低pBOO大鼠膀胱重量及膀胱与体重的比值(均P<0.05); 显著逆转膀胱组织低顺应性和提高膀胱肌张力(均P<0.05); 明显抑制大鼠膀胱组织中炎症细胞浸润; GE还能显著降低MDA含量、增加SOD及GSH-Px活性(均P<0.05); 显著逆转pBOO模型大鼠Nrf2/NF-κB通路相关蛋白表达及基因mRNA表达水平(均P<0.05)。结论:GE可以明显改善pBOO模型大鼠的膀胱功能障碍,其可能与调控Nrf2/NF-κB通路有关。
Abstract:
Objective:To investigate the therapeutic effects and potential mechanisms of geniposide(GE)on the rat model of partial bladder outlet obstruction(pBOO).Methods:Rats were randomly divided into five groups(n=6):Sham group,pBOO group,and GE 20,40 and 80 mg/kg groups.Four weeks after pBOO surgery,GE was administered by gavage once a day for 2 weeks.The effects of GE on bladder muscle tone and bladder urodynamic characteristics of rats were detected.HE staining was used to evaluate the improvement effect of GE on inflammatory cell infiltration in the bladder tissue.The content of malondialdehyde(MDA),the activity of superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px)were detected according to the manufacturer's instructions.The Nrf2/NF-κB pathway-related protein and mRNA levels were detected by western blot and quantitative real-time PCR.Results:GE 40 and 80 mg/kg groups could significantly reduce the bladder weight and the ratio of the bladder to body weight(all P<0.05).GE significantly reversed the low compliance of bladder tissue and improved bladder muscle tone(all P<0.05).GE significantly inhibited the infiltration of inflammatory cells in the bladder tissue,and reduced the MDA content,and increase SOD and GSH-Px activities(all P<0.05).GE significantly reversed the Nrf2/NF-κB pathway-related protein and mRNA levels(all P<0.05).Conclusion:GE can significantly improve bladder dysfunction in a rat model of pBOO,and its mechanism may be related to the regulation of the Nrf2/NF-κB pathway.
