摘要:
目的:探讨黄芪多糖联合替加环素对白细胞减少性鲍曼不动杆菌感染肺炎大鼠的疗效及机制。方法:实验开始前,将75只大鼠分为五组,即空白对照组、模型组、黄芪多糖组、替加环素组、黄芪多糖联合替加环素组。除空白对照组,其余四组均进行造模,动物造模后,黄芪多糖组给予1 ml/100 g黄芪多糖,替加环素给予1 ml/100 g替加环素,黄芪多糖联合替加环素组给予1 ml/100 g黄芪多糖+1 ml/100 g替加环素,正常对照组和模型组大鼠给予等量的0.9%氯化钠溶液进行灌胃,五组均连续灌胃给药7 d。分析五组大鼠的造模结果、精神活动状况及大鼠的生存时间、湿干比、白细胞计数、细菌含量、血清细胞因子、检测Toll样受体4(TLR4)、核转录因子-κB(NF-κB)相对表达量。结果:75只大鼠造模成功率86.67%(65/75)。所有大鼠给药7 d内均存活,黄芪多糖联合替加环素组的生存时间明显较黄芪多糖组、替加环素组、模型组高,黄芪多糖组、替加环素组明显较模型组高(均P<0.05),黄芪多糖组与替加环素组比较无统计学意义(P>0.05)。给药第1天,模型组、黄芪多糖组、替加环素组、黄芪多糖联合替加环素组的湿干比、细菌含量、基质金属蛋白酶-9(MMP-9)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、TLR4、NF-κB明显较空白对照组高,白细胞计数明显较低(均P<0.05); 模型组、黄芪多糖组、替加环素组、黄芪多糖联合替加环素组组间的湿干比、白细胞计数、细菌含量、MMP-9、IL-6、TNF-α、TLR4、NF-κB比较无统计学意义(均P>0.05); 给药第3天、第5天、第7天,模型组的湿干比、细菌含量、MMP-9、IL-6、TNF-α、TLR4、NF-κB明显较空白对照组、黄芪多糖组、替加环素组、黄芪多糖联合替加环素组高,白细胞计数明显较低; 黄芪多糖组、替加环素组的湿干比、细菌含量、MMP-9、IL-6、TNF-α、TLR4、NF-κB明显较空白对照组、黄芪多糖联合替加环素组高,白细胞计数明显较低; 黄芪多糖联合替加环素组的湿干比、细菌含量、MMP-9、IL-6、TNF-α、TLR4、NF-κB明显较空白对照组高,白细胞计数明显较低(均P<0.05); 黄芪多糖组、替加环素组的湿干比、白细胞计数、细菌含量、MMP-9、IL-6、TNF-α、TLR4、NF-κB比较无统计学意义(均P>0.05)。结论:黄芪多糖联合替加环素对白细胞减少性鲍曼不动杆菌感染肺炎大鼠有一定疗效,可能与其可通过TLR4/NF-κB通路改善炎症水平有关。
Abstract:
Objective:To investigate the curative effect and mechanism of astragalus polysaccharide combined with tigacycline on leukopenic acinetobacter baumannii pneumonia in rats with leukopenia.Methods:Before the experiment,75 rats were divided into 5 groups,including blank control group,model group,astragalus polysaccharide group,tegacycline group and astragalus polysaccharide combined with tigacycline group.Except blank control group,the other 4 groups were modeled.After animal modeling,astragalus polysaccharide group was given 1 ml/100 g astragalus polysaccharide,tigecycline was given 1 ml/100 g tigecycline,and astragalus polysaccharide combined with tigecycline group was given 1 ml/100 g astragalus polysaccharide +1 ml/100 g tigecycline.Normal control group and model group were given the same amount of normal saline intragastric administration,the 5 groups were given continuous intragastric administration for 7 days.The results of model building,mental activity,survival time,wet-dry ratio,white blood cell count,bacterial content,serum cytokines,and relative expression levels of TLR4 and NF-κB of the 5 groups were analyzed.Results:The success rate of 75 rats was 86.67%(65/75).All rats survived within 7 days of administration.The survival time of APS combined with tigacycline group was significantly higher than that of APS group,tigacycline group and model group,while APS group and tigacycline group were significantly higher than model group(all P<0.05).There was no statistical significance between APS group and tigacycline group(P>0.05).On day 1 after administration,the wet-dry ratio,bacterial content,MMP-9,IL-6,TNF-α,TLR4 and NF-κB of model group,astragalus polysaccharide group,tigacycline group and astragalus polysaccharide combined tigacycline group were significantly higher than those of blank control group,and the WBC count was significantly lower(all P<0.05).There was no statistical significance in wet to dry ratio,white blood cell count,bacterial content,MMP-9,IL-6,TNF-α,TLR4 and NF-κB among model group,astragalus polysaccharide group,tigacycline group,and astragalus polysaccharide combined tigacycline group(all P>0.05).At day 3,day 5 and day 7,the wet-dry ratio,bacterial content,MMP-9,IL-6,TNF-α,TLR4,NF-κB in model group were significantly higher than those in the control group,astragalus polysaccharide group,tigacycline group and astragalus polysaccharide combined with tigacycline group,and the WBC count was significantly lower.The wet-dry ratio,bacterial content,MMP-9,IL-6,TNF-α,TLR4 and NF-κB of APS and tigacycline groups were significantly higher than those of blank control group and APS combined with tigacycline group,and the WBC count was significantly lower.The wet-dry ratio,bacterial content,MMP-9,IL-6,TNF-α,TLR4 and NF-κB of APS combined with tigacycline group were significantly higher than those of blank control group,and the WBC count was significantly lower(all P<0.05).There were no significant differences in wet to dry ratio,white blood cell count,bacterial content,MMP-9,IL-6,TNF-α,TLR4 and NF-κB between APS group and tigacycline group(all P>0.05).Conclusion:Astragalus polysaccharide combined with tigacycline has a certain effect on rats infected with pneumonia caused by leukopenic acinetobacter baumannii leukopenia,which may be related to the improvement of inflammation level through TLR4/NF-κB pathway.
