摘要:
目的:探讨白头翁皂苷A对鼻咽癌细胞恶性生物学行为的影响及分子机制。方法:体外培养人鼻咽上皮细胞NP69和鼻咽癌细胞系(HK-1、HONE1、CNE2、SUNE1、C666-1),实时荧光定量PCR(RT-qPCR)检测细胞中circ-0001742表达水平; 将C666-1细胞分为si-circ-0001742组、si-NC组、对照组、白头翁皂苷A-L、M、H组、白头翁皂苷A-H+pcDNA-circ-0001742组; 四甲基偶氮唑盐比色法(MTT)检测细胞存活率,流式细胞术检测细胞凋亡,Transwell检测细胞迁移数,克隆形成实验检测细胞克隆数,蛋白质印迹法检测蛋白表达。结果:鼻咽癌细胞(HK-1、HONE1、CNE2、SUNE1、C666-1)中circ-0001742表达水平升高(均P<0.05)。沉默circ-0001742后,C666-1细胞增殖抑制率、细胞凋亡率以及活化的半胱天冬氨酸蛋白酶3(Cleaved caspase-3)表达水平升高,C666-1细胞迁移和克隆细胞数减少(均P<0.05)。不同浓度白头翁皂苷A处理后,C666-1细胞增殖抑制率、细胞凋亡率以及Cleaved caspase-3表达水平升高,C666-1细胞迁移和克隆细胞数减少,circ-0001742表达水平降低,呈浓度依赖性(均P<0.05)。circ-0001742过表达可逆转白头翁皂苷A对C666-1细胞的抑制作用。结论:白头翁皂苷A通过下调circ-0001742抑制鼻咽癌C666-1细胞恶性生物学行为。
Abstract:
Objective:To explore the effects of pulsatilla saponin A on the malignant biological behavior of nasopharyngeal carcinoma cells and its molecular mechanism.Methods:Human nasopharyngeal epithelial cells NP69 and nasopharyngeal carcinoma cell lines(HK-1,HONE1,CNE2,SUNE1,C666-1)were cultured in vitro,and the expression level of circ-0001742 was detected by RT-qPCR.C666-1 cells were divided into si-circ-0001742 group,si-NC group,control group,Pulsatilla saponin A-L,M,H group,Pulsatilla saponin A-H+pcDNA-circ-0001742 group.The cell survival rate was determined by MTT.Apoptosis was detected by flow cytometry.Transwell was used to detect the number of cell migration.The clone formation test was used to detect the number of cell clones.Western blot was used to detect protein expression.Results:The expression levels of circ-0001742 in nasopharyngeal carcinoma cells(HK-1,HONE1,CNE2,SUNE1,C666-1)increased(all P<0.05).After circ-0001742 silenced,C666-1 cell proliferation inhibition rate,cell apoptosis rate,Cleaved caspase-3 expression level increased,C666-1 cell migration and number of cloned cells decreased(all P<0.05).After treatment with different concentrations,C666-1 cell proliferation inhibition rate,cell apoptosis rate,Cleaved caspase-3 expression level increased,C666-1 cell migration and number of cloned cells decreased,and circ-0001742 expression level decreased in a concentration-dependent manner(all P<0.05).Overexpression of circ-0001742 reversed the inhibitory effect of pulsatilla saponin A on C666-1 cells.Conclusion:Pulsatilla saponin A inhibits the malignant biological behavior of nasopharyngeal carcinoma C666-1 cells by down-regulating circ-0001742.
