摘要:
目的:检测结肠癌患者血清微小RNA-193a-5p(miR-193a-5p)和微小RNA-139-5p(miR-139-5p)表达水平,分析其对预后的影响。方法:选取结肠癌患者109例为结肠癌组,选取同期体检健康者100例为健康组。收集两组临床资料,采用实时荧光定量PCR检测血清miR-193a-5p和miR-139-5p的表达。分析miR-193a-5p和miR-139-5p表达与结肠癌患者临床病理特征的关系。采用 Cox 风险比例模型分析结肠癌患者预后的影响因素。绘制Kaplan-Meier曲线进行生存分析。结果:结肠癌组miR-193a-5p和miR-139-5p表达明显低于健康组(均P<0.001)。结肠癌患者血清miR-193a-5p和miR-139-5p表达与性别、年龄、病理分型、肿瘤部位、肿瘤直径无明显相关性(均P>0.05 ),与临床分期、分化程度、局部浸润深度和淋巴结转移具有明显相关性(均P<0.001)。Cox风险比例模型分析结果显示,临床分期、分化程度、局部浸润深度、淋巴结转移、miR-193a-5p和miR-139-5p是结肠癌患者预后的影响因素(均P<0.05)。miR-193a-5p低表达组患者5年总生存率明显低于miR-193a-5p高表达组,miR-139-5p低表达组患者5年总生存率明显低于miR-139-5p高表达组(均P<0.05)。结论:血清miR-193a-5p和miR-139-5p在结肠癌患者中表达均明显降低,可能在结肠癌中发挥着抑癌基因的作用。检测血清中miR-193a-5p和miR-139-5p的表达水平有助于结肠癌的诊断及预后评估,可能成为结肠癌早期诊断和预后判断的分子标志物。
Abstract:
Objective:To detect the expression levels of serum microRNA-193a-5p(miR-193a-5p)and microRNA-139-5p(miR-139-5p)in patients with colon cancer,and to analyze their effects on prognosis.Methods:A total of 109 patients with colon cancer were selected as the colon cancer group,and 100 healthy people in the same period were selected as the healthy group.The clinical data of the two groups were collected,and the expression levels of serum miR-193a-5p and miR-139-5p were detected by real-time fluorescence quantitative PCR.The relationship between miR-193a-5p,miR-139-5p expression and clinicopathological characteristics of colon cancer patients was analyzed.Cox risk proportional model was used to analyze the prognostic factors of colon cancer patients.Kaplan-Meier curves were drawn for survival analysis.Results:The expression levels of miR-193a-5p and miR-139-5p in the colon cancer group were significantly lower than those in the healthy group(both P<0.001).The expression of serum miR-193a-5p and miR-139-5p in patients with colon cancer were not significantly correlated with gender,age,pathological type,tumor location and tumor diameter(all P>0.05),but were significantly correlated with clinical stage,differentiation degree,local invasion depth and lymph node metastasis(all P<0.001).Cox risk proportional model analysis showed that clinical stage,degree of differentiation,depth of local invasion,lymph node metastasis,miR-193a-5p and miR-139-5p were prognostic factors for colon cancer patients(all P<0.05).The 5-year overall survival rate of patients in the miR-193a-5p low expression group was significantly lower than that in the miR-193a-5p high expression group,and the 5-year overall survival rate of patients in the miR-139-5p low expression group was significantly lower than that in the miR-139-5p high expression group(both P<0.05).Conclusion:The expressions of serum miR-193a-5p and miR-139-5p are significantly decreased in patients with colon cancer,which may play a role as tumor suppressor genes in colon cancer.Detection of serum miR-193a-5p and miR-139-5p expression levels is helpful for the diagnosis and prognosis evaluation of colon cancer,and may become molecular markers for early diagnosis and prognosis of colon cancer.
