摘要:
目的:探讨长链非编码RNA(lncRNA)C16orf89在胃癌组织及细胞系中的表达,观察lncRNA C16orf89过表达对胃癌细胞增殖、侵袭的影响及其作用机制。方法:应用基因表达谱交互分析(GEPIA)数据库在线分析lncRNA C16orf89在胃癌组织中的表达。应用Oncolnc数据库在线分析lncRNA C16orf89表达量与胃癌患者总生存期的关系。采用实时定量聚合酶链反应(qRT-PCR)检测lncRNA C16orf89在胃癌细胞系中的表达。分别将阴性对照质粒和lncRNA C16orf89过表达质粒转染至lncRNA C16orf89表达最低的胃癌细胞系,分别定义为NC组和lncRNA C16orf89组。采用噻唑蓝(MTS)和Transwell实验检测lncRNA C16orf89过表达对胃癌细胞增殖及侵袭的影响。采用在线生物信息学网站分析可能与lncRNA C16orf89结合的微小RNA(miRNA)。采用双荧光素酶报告载体实验检测lncRNA C16orf89与miR-95-3p的相互作用。采用qRT-PCR检测lncRNA C16orf89过表达对miR-95-3p表达的影响。采用Western blot检测NC组和lncRNA C16orf89组细胞磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)信号通路蛋白的表达。结果:胃癌组织中lncRNA C16orf89 表达量明显低于癌旁组织(P<0.01)。与lncRNA C16orf89低表达的胃癌患者相比,lncRNA C16orf89高表达的胃癌患者总生存期较长(P<0.01)。胃癌细胞系中lncRNA C16orf89表达量明显低于GES-1细胞,且AGS细胞中lncRNA C16orf89表达量最低(均P<0.01)。与NC组相比,lncRNA C16orf89过表达显著抑制AGS细胞的增殖和侵袭能力(均P<0.05)。lncRNA C16orf89能够靶向结合miR-95-3p。与NC组比较,lncRNA C16orf89过表达明显抑制miR-95-3p的表达,lncRNA C16orf89组细胞中PI3K/AKT信号通路蛋白磷酸化磷脂酰肌醇3-激酶(p-PI3K)、磷酸化蛋白激酶B(p-AKT)、磷酸化雷帕霉素靶蛋白(p-mTOR)、细胞周期蛋白D1(CCDN1)表达降低,磷酸酯酶与张力蛋白同源物(PTEN)蛋白表达增加(均P<0.01)。结论:胃癌中lncRNA C16orf89呈低表达状态,其通过竞争结合miR-95-3p干扰PI3K/AKT信号通路转导,进而抑制胃癌细胞的增殖及侵袭。
Abstract:
Objective:To research the expression of long non-coding RNA(lncRNA)C16orf89 in gastric cancer tissues and cell lines,and to observe the effect of lncRNA C16orf89 overexpression on the proliferation and invasion of gastric cancer cells and its mechanism.Methods:The expression of lncRNA C16orf89 in gastric cancer tissues was analyzed online by using GEPIA database.The Oncolnc database was used to analyze the relationship between the expression of lncRNA C16orf89 and the overall survival of patients with gastric cancer.The expression of lncRNA C16orf89 in gastric cancer cell lines was detected by qRT-PCR.Negative control plasmid and lncRNA C16orf89 overexpression plasmid were transfected into gastric cancer cell lines with the lowest expression of lncRNA C16orf89,which were defined as NC group and lncRNA C16orf89 group,respectively.MTT assay and Transwell assay were used to detect the effect of lncRNA C16orf89 overexpression on the proliferation and invasion of gastric cancer cells.The online bioinformatics website was used to analyze miRNAs that might bind to lncRNA C16orf89,and the interaction between lncRNA C16orf89 and miR-95-3p was detected by dual-luciferase reporter vector assay.The effect of lncRNA C16orf89 overexpression on the expression of miR-95-3p was detected by qRT-PCR.Western blot was used to detect the expression of PI3K/AKT signaling pathway proteins in the cells of NC group and lncRNA C16orf89 group.Results:The expression of lncRNA C16orf89 in gastric cancer tissues was significantly lower than that in adjacent tissues(P<0.01).Compared with gastric cancer patients with low expression of lncRNA C16orf89,patients with gastric cancer with high expression of lncRNA C16orf89 had longer overall survival(P<0.01).The expression level of lncRNA C16orf89 in gastric cancer cell lines was significantly lower than that in GES-1 cells,and the expression level of lncRNA C16orf89 in AGS cells was the lowest(all P<0.01).Compared with the NC group,lncRNA C16orf89 overexpression significantly inhibited the proliferationand invasion ability of AGS cells(all P<0.05).LncRNA C16orf89 could target and bind miR-95-3p.Compared with the NC group,the overexpression of lncRNA C16orf89 significantly inhibited the expression of miR-95-3p,and the expression of PI3K/AKT signaling pathway proteins p-PI3K,p-AKT,p-mTOR,CCDN1 in the lncRNA C16orf89 group decreased,and the expression of PTEN protein increased(all P<0.01).Conclusion:The expression of lncRNA C16orf89 in gastric cancer is low,and it interferes with the transduction of PI3K/AKT signaling pathway by competing with miR-95-3p,thereby inhibiting the proliferation and invasion of gastric cancer cells.
