摘要:
目的:探讨脂多糖(LPS)诱导的急性炎症小鼠模型血清促炎因子、炎性因子表达情况。方法:将120只健康雄性ICR小鼠随机分为空白组、A组、B组和C组,各30只。分别给予A组、B组和C组小鼠经腹腔注射2、4 mg/kg和6 mg/kg LPS 0.9%氯化钠溶液,给予空白组小鼠等体积0.9%氯化钠溶液。建模成功后断头处死小鼠采集血液样本后检测血清中促炎因子[肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、IL-6、IL-18]、抑炎因子(IL-4、IL-10、IL-13),取肾脏和肺脏采用TUNEL染色法检测细胞凋亡情况,采用Western blot技术检测肾脏和肺脏组织中核因子-κB(NF-κB)、磷酸化NF-κB(pNF-κB)、Toll样受体4(TLR4)相对表达水平。结果:A组、B组和C组小鼠血清TNF-α、IL-1β、IL-6、IL-18、IL-4、IL-10、IL-13水平以及肺和肾脏组织细胞凋亡比例均显著高于空白组(均P<0.05)。随着LPS注射浓度增加,小鼠血清TNF-α、IL-1β、IL-6、IL-18、IL-4、IL-10、IL-13水平以及肺和肾脏组织细胞凋亡比例呈显著升高趋势(均P<0.05)。Pearson分析结果显示血清TNF-α、IL-1β、IL-6、IL-18、IL-4、IL-10、IL-13水平与肺组织和肾脏组织细胞凋亡比例均呈显著正相关性(均P<0.05)。各组小鼠肺组织和肾脏组织pNF-κB和TLR4相对表达水平随LPS注射浓度增加呈显著升高趋势(均P<0.05)。结论:LPS诱导急性炎症小鼠血清促炎因子和抑炎因子均呈高表达,可造成肺、肾脏损伤,这可能与NF-κB信号通路激活有关。
Abstract:
Objective:To investigate the expression of pro-inflammatory factors and anti-inflammatory factors in serum of mice with acute inflammation induced by lipopolysaccharide(LPS).Methods:A total of 120 healthy male ICR mice were randomly divided into blank group,group A,group B and group C,with 30 mice in each group.The mice in Group A,Group B and Group C were injected with 2 mg/kg,4 mg/kg and 6 mg/kg LPS saline solution by intraperitoneal injection respectively,and the mice in the blank group were given the same volume of 0.9% sodium chloride solution.After successful modeling,the mice were decapitated,and blood samples were collected to detect pro-inflammatory factors(TNF-α,IL-1β,IL-6,IL-18),anti-inflammatory factors(IL-4,IL-10,IL-13).TUNEL staining was used to detect apoptosis of kidney and lung cells,and Western blot was used to detect relative expression levels of NF-κB,pNF-κB and TLR4 in kidney and lung tissues.Results:The levels of serum TNF-α,IL-1β,IL-6,IL-18,IL-4,IL-10,IL-13 and the percentage of apoptosis in lung and kidney tissues of mice in group A,B and C were significantly higher than those in the blank group(all P<0.05).With the increase of LPS injection concentration,the levels of serum TNF-α,IL-1β,IL-6,IL-18,IL-4,IL-10,IL-13 and the percentage of apoptosis in lung and kidney tissues of mice increased significantly(all P<0.05).Pearson analysis showed that the levels of serum TNF-α,IL-1β,IL-6,IL-18,IL-4,IL-10,IL-13 were significantly positively correlated with the percentage of apoptosis in lung and kidney tissues(all P<0.05).The relative expression levels of pNF-κB and TLR4 in lung and kidney tissues of mice in each group increased significantly with the increase of LPS injection concentration(all P<0.05).Conclusion:LPS-induced acute inflammation in mice show high expression of serum pro-inflammatory factors and anti-inflammatory factors,which may cause lung and kidney damage,which may be related to NF-κB signal path activation.
