目的:探究微小RNA-325-3p(miR-325-3p)与叉头框蛋白M1(FOXM1)在胃腺癌中的表达,分析miR-325-3p与FOXM1的靶向关系。方法:选择人胃腺癌MGC-803、SGC-7901细胞系,分别构建miR-325-3p mimic组、miR-325-3p inhibitor组,并设立空白对照组,应用Western blot法检测FOXM1蛋白的表达,应用双荧光素酶报告基因实验验证miR-325-3p与FOXM1的靶向关系。收集确诊为胃腺癌并行根治术的42例患者的肿瘤组织和距肿物边缘>2 cm的癌旁正常胃黏膜组织,应用实时荧光定量PCR法(qRT-PCR)检测miR-325-3p的表达,应用免疫组化染色(EnVision法)检测组织中FOXM1的表达。结果:胃癌MGC-803和SGC-7901细胞系miR-325-3p mimic组FOXM1蛋白表达明显低于空白对照组,miR-325-3p inhibitor组FOXM1蛋白表达明显高于空白对照组(均P<0.05)。双荧光素酶报告基因实验显示,miR-325-3p与FOXM1具有靶向关系。胃腺癌组织中miR-325-3p相对表达量明显低于癌旁正常胃黏膜组织,FOXM1阳性率明显高于癌旁正常胃黏膜组织(均P<0.05)。miR-325-3p和FOXM1在胃腺癌不同肿瘤最大径、浸润深度方面比较差异有统计学意义(均P<0.05)。Pearson相关性分析显示,miR-325-3p与FOXM1呈负相关(r=-0.630,P=0.027)。结论:胃腺癌中miR-325-3p与FOXM1异常表达,参与肿瘤的形成和进展。miR-325-3p与FOXM1具有靶向调控关系。
Abstract:
Objective:To investigate the expression of microRNA-325-3p(miR-325-3p)and forkhead box M1(FOXM1)in gastric adenocarcinoma,and to analyze the targeting relationship between miR-325-3p and FOXM1.Methods:Gastric cancer cell lines MGC-803 and SGC-7901 were selected to construct miR-325-3p mimic group,miR-325-3p inhibitor group and blank control group respectively,and the expression of FOXM1 was detected by Western blot,and targeting relationship between miR-325-3p and FOXM1 was observed by dual luciferase reporter gene assay.Tumor tissues and adjacent normal gastric mucosa tissues >2 cm from tumor margin were collected from 42 patients who were diagnosed with gastric adenocarcinoma and underwent radical operation,and the expression of miR-325-3p was detected by qRT-PCR and FOXM1 was detected by immunohistochemical staining(EnVision method).Results:The expression of FOXM1 was significantly lower in miR-325-3p mimic group than that in blank control group,and the expression of FOXM1 was significantly higher in miR-325-3p inhibitor group than that in blank control group of both gastric cancer cell lines MGC-803 and SGC-7901(all P<0.05).Dual luciferase reporter gene assay showed that miR-325-3p had a targeting relationship with FOXM1.The relative expression level of miR-325-3p in gastric adenocarcinoma tissues was significantly lower than that in adjacent normal gastric mucosa tissues,and the positive rate of FOXM1 was significantly higher than that in adjacent normal gastric mucosa tissues(all P<0.05).There were statistically significant differences between miR-325-3p and FOXM1 in the maximum diameter and invasion depth of different gastric adenocarcinoma tumors(all P<0.05).Pearson correlation analysis showed that miR-325-3p was negatively correlated with FOXM1(r=-0.630,P=0.027).Conclusion:The abnormal expression of miR-325-3p and FOXM1 in gastric adenocarcinoma is involved in the formation and progression of tumor.MiR-325-3p has a targeted regulatory relationship with FOXM1.
