摘要:
目的:探讨紫草素对慢性髓系白血病K562细胞的作用及其可能机制。方法:体外培养慢性髓系白血病K562细胞,分组为对照组和紫草素10 μmol/L组、紫草素30 μmol/L组、紫草素50 μmol/L组,分别培养12、24、48、72 h后,MTT法检测细胞增殖抑制率,Annexin V FITC/PI染色结合流式细胞仪检测细胞凋亡率,同时用MTT法观察紫草素对K562细胞生长的影响; Western blot检测细胞内PTEN、p-PI3K和p-Akt蛋白表达。结果:紫草素能有效抑制K562细胞增殖,促进细胞凋亡,上调细胞PTEN蛋白表达量,下调p-PI3K和p-Akt蛋白表达量,各浓度间差异具有统计学意义(均P<0.05)。结论:紫草素能通过调控PTEN、p-PI3K、p-Akt信号通路抑制K562细胞的增殖,促进其凋亡。
Abstract:
Objective:To study the effect of shikonin on K562 cells and its possible mechanism.Methods:K562 cells were cultured in vitro and divided into control group,shikonin 10 μmol/L group,shikonin 30 μmol/L group and shikonin 50 μmol/L group.After 12,24,48 and 72 hours of culture,the inhibition rate of cell proliferation was detected by MTT assay.The apoptosis rate of K562 cells was detected by Annexin V FITC/PI staining combined with flow cytometry,and the effect of shikonin on the growth of K562 cells was observed by MTT assay.The protein expressions of PTEN,p-PI3K and p-Akt were detected by Western blot.Results:Shikonin could effectively inhibit the proliferation of K562 cells,promote the apoptosis of cells,up-regulate the protein expression of PTEN,down-regulate the protein expression of p-Pi3k and p-Akt,with statistically significant differences between different concentrations(all P<0.05).Conclusion:Shikonin can inhibit the proliferation and promote the apoptosis of AK562 cells by regulating PTEN,p-Pi3k,p-Akt signaling pathway.
