摘要:
目的:探讨麦冬多糖对衰老大鼠皮肤形态及5-羟色胺(5-HT)表达变化的影响。方法:将30只SD大鼠分为对照组、模型组和麦冬多糖组,每组10只。使用酶联免疫吸附法试剂盒测定大鼠脑组织中5-HT、皮肤抗氧化活性物质和细胞因子水平。使用HE染色观察皮肤上皮细胞病理损伤。通过Masson染色确定皮肤胶原蛋白的分布。结果:模型组5-HT水平较对照组降低,麦冬多糖组5-HT水平较模型组升高(均P<0.05)。模型组皮肤匀浆组织抗氧化活性物质超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)水平较对照组降低,麦冬多糖组SOD、CAT和GSH-Px水平较模型组升高(均P<0.05)。模型组白细胞介素-6(IL-6)、IL-8和核因子-κB(NF-κB)水平较对照组升高,麦冬多糖组IL-6、IL-8和NF-κB水平较模型组降低(均P<0.05)。模型组基质金属蛋白酶-1(MMP-1)和粒细胞-巨噬细胞集落刺激因子(GM-CSF)水平较对照组升高,麦冬多糖组MMP-1和GM-CSF水平较模型组降低(均P<0.05)。与对照组相比,模型组可观察到表皮结构不完整、真皮胶原纤维少而变细、真皮胶原纤维排列紊乱、真皮炎症细胞浸润等病理变化,而麦冬多糖组可显著缓解模型组的病理变化。模型组皮肤纤维组织表达面积百分比和Ⅰ/Ⅲ型胶原蛋白比值较对照组降低,麦冬多糖组皮肤纤维组织表达面积百分比和Ⅰ/Ⅲ型胶原蛋白比值较模型组升高(均P<0.05)。结论:麦冬多糖通过发挥抗氧化活性改善因衰老引起的5-HT表达下降,抑制细胞凋亡,延缓皮肤衰老。
Abstract:
Objective:To investigate the effect of Ophiopogon japonicus polysaccharide on the changes of skin morphology and serotonin(5-HT)expression in aging rats.Methods:A total of 30 SD rats were divided into control group,model group and Ophiopogon japonicus polysaccharide group,with 10 rats in each group.The levels of 5-HT,skin antioxidant active substances and cytokines in rat brain tissue were determined by using enzyme-linked immunosorbent assay kits.The pathological damage of skin epithelial cells was observed by HE staining,and the distribution of skin collagen was determined by Masson staining.Results:The level of 5-HT in the model group was lower than that in the control group,and the level of 5-HT in the Ophiopogon polysaccharide group was higher than that in the model group(all P<0.05).The levels of antioxidant active substances SOD,CAT and GSH-Px in the skin homogenate of the model group were lower than those of the control group,and the levels of SOD,CAT and GSH-Px in the Ophiopogon polysaccharide group were higher than those of the model group(all P<0.05).The levels of IL-6,IL-8 and NF-κB in the model group were higher than those in the control group,and the levels of IL-6,IL-8 and NF-κB in the Ophiopogon polysaccharide group were lower than those in the model group(all P<0.05).The levels of matrix metalloproteinase-1(MMP-1)and granulocyte-macrophage colony-stimulating factor(GM-CSF)in the model group were higher than those in the control group,and the levels of MMP-1 and GM-CSF in the Ophiopogon japonicus polysaccharide group were lower than those in the model group(all P<0.05).Compared with the control group,pathological changes such as incomplete epidermal structure,fewer and thinner dermal collagen fibers,disordered dermal collagen fibers,and infiltration of dermal inflammatory cells were observed in the model group,while the Ophiopogon polysaccharide group significantly alleviated the pathological tissue in the model group.Compared with the control group,the percentage of skin fibrous tissue expression and type Ⅰ/Ⅲ collagen in the model group decreased,while the percentage of skin fibrous tissue expression and type Ⅰ/Ⅲ collagen in the Ophiopogon polysaccharide group increased in comparison with the model group(all P<0.05).Conclusion:Ophiopogon japonicus polysaccharide can improve the expression of 5-HT caused by aging,inhibit apoptosis and delay skin aging by exerting antioxidant activity.
