摘要:
目的:研究辅酶Q10(CoQ10)对脑出血(ICH)小鼠的神经功能、周围组织水肿、炎症因子水平和细胞凋亡的影响。方法:将54只C57WT小鼠随机分为Sham组、ICH组、CoQ10组,采用 Ⅳ型胶原酶注射到基底神经节的方法建立脑出血模型,CoQ10组小鼠在脑出血模型建立6 h后,给予CoQ10灌胃处理。于造模后7 d时对所有小鼠进行mNSS评分评估其神经功能,并用公式计算小鼠脑水肿程度; 于3 d时采用酶联免疫吸附试验检测炎症因子白介素6(IL-6)及肿瘤坏死因子-α(TNF-α)的表达水平,TUNEL染色法检测细胞凋亡水平。结果:CoQ10治疗降低了脑出血小鼠造模7 d后的mNSS评分(P<0.05),脑组织水含量计算的结果也显示ICH组脑水肿程度加重(P<0.001),而CoQ10组可减轻小鼠的脑水肿(P<0.01)。与ICH组相比,经CoQ10处理可以显著降低炎症因子的表达水平(P<0.01)及减少细胞凋亡水平(P<0.001)。结论:CoQ10可减轻脑出血小鼠神经功能障碍、降低脑水肿程度、减少炎症因子产生、改善细胞凋亡。
Abstract:
Objective:To study the effects of coenzyme Q10(CoQ10)on nerve function,surrounding tissue edema,inflammatory factor levels and apoptosis of intracerebral hemorrhage(ICH)mice.Methods:Fifty-four C57 WT mice were randomly divided into Sham group,ICH group and CoQ10 group.ICH group and CoQ10 group were injected with type Ⅳ collagenase into basal ganglia to establish ICH model.CoQ10 group was given CoQ10 intragastric treatment 6 hours after the establishment of ICH model.At 7 days after modeling,all mice were evaluated by mNSS score,and the degree of cerebral edema was calculated by formula.At 3 days,the expression levels of inflammatory factors were detected by enzyme-linked immunosorbent assay,and the apoptosis level was detected by TUNEL staining.Results:CoQ10 treatment reduced the mNSS score(P<0.05),the calculation results of brain water content also showed that the degree of cerebral edema increased in the ICH group(P<0.001),while CoQ10 reduced cerebral edema in mice(P<0.01).Compared with ICH group,CoQ10 could significantly reduce the expression level of inflammatory factors and decrease apoptosis levels(all P<0.001).Conclusion:CoQ10 can relieve the neurological dysfunction,reduce the degree of cerebral edema,reduce the production of inflammatory factors and improve cell apoptosis in mice with ICH.
