摘要:
目的:探索柚皮素对创伤性颅脑损伤后炎性反应的作用以及机制。方法:50只小鼠随机分为Sham组、TBI组、柚皮素低、中、高剂量组,每组10只,采用限制性皮层损伤造模,造模1 h后,低剂量、中剂量、高剂量组小鼠每日腹腔注射柚皮素,共7 d; Sham组和TBI组小鼠仅注射同等剂量柚皮素溶剂。采用mNSS评分进行神经功能评估、采用酶联免疫吸附实验检测脑组织中相关炎症因子的表达变化情况,采用Tunel法和免疫荧光法分别检测神经元细胞的凋亡率和小胶质细胞的活化状态。结果:相较于Sham组,TBI组小鼠mNSS评分明显上调(P<0.05),脑组织中细胞凋亡率以及炎症因子白细胞介素(IL)-1β、IL-6、肿瘤坏死因子(TNF-α)的表达水平均呈现显著升高趋势(均P<0.05),同时小胶质细胞活化的比例也有明显增加; 给药组小鼠mNSS评分显著下降(P<0.05),脑组织中细胞凋亡率以及炎症因子IL-1β、IL-6、TNF-α的表达水平均有明显降低(均P<0.05),同时降低了Iba1阳性细胞的数量以及小胶质细胞的活化状态,并且以上结果均呈剂量依赖性。结论:柚皮素可以缓解TBI后炎性反应,减少细胞凋亡,改善TBI小鼠的预后情况,可能与抑制小胶质细胞的活化有关。
Abstract:
Objective:To explore the role and mechanism of naringenin in inflammatory response after traumatic brain injury.Methods:A total of 50 mice were randomly divided into Sham group,TBI group,naringenin low-dose group,naringenin medium-dose group and naringenin high-dose group,with 10 mice in each group.Models were established with restricted cortex injury.One hour after modeling,mice in naringenin low-dose group,naringenin medium-dose group and naringenin high-dose group were injected with naringenin intraperitoneally daily for 7 days.Mice of Sham group and TBI group were injected with equal amount of naringenin solvent.The neurological function of mice was evaluated by mNSS score.The expression levels of related inflammatory factors in brain tissue were detected by EELISA.TUNEL method was used for sensing the neuron apoptosis,and the activation state of microglia was monitored by immunofluorescence.Results:Compared with Sham group,the mNSS score in TBI group was significantly increased(P<0.05),the apoptosis rate and the expression levels of IL-1β,IL-6 and TNF-α in brain tissue were significantly increased(all P<0.05),and the activation rate of microglia was also significantly increased.Compared with TBI group,the mNSS score of mice in administration group descended significantly(P<0.05),the apoptosis rate of nerve cells and the expression levels of inflammatory factors were significantly down-regulated(all P<0.05),and the number of Iba1 positive cells and the activation status of microglia were decreased,and The above results were dose-dependent.Conclusion:Naringenin can alleviate the inflammatory response after TBI,reduce apoptosis and improve the prognosis of TBI mice,which may be related to inhibiting the activation of microglia.
