摘要:
目的:探究依那西普对银屑病大鼠模型固有淋巴细胞亚群分型的影响。方法:30只6周龄雄性Lewis大鼠分为对照组、模型组和依那西普组,每组10只。通过咪喹莫特诱导银屑病大鼠模型,通过银屑病皮损严重程度指数对大鼠背部分型进行评分,通过流式细胞术分析大鼠外周血ILC1、ILC2和ILC3的水平,通过蛋白印迹检测转录因子T-bet、GATA3、RORγt、Caspase-3、Caspase-14、丝聚蛋白和外皮蛋白的蛋白表达,通过RT-PCR检测大鼠银屑病皮肤组织炎性细胞因子(IFN-γ、IL-5、IL-13、IL-17和IL-22)的mRNA表达。结果:模型组红斑、鳞屑和厚度评分较对照组升高(均P<0.05),依那西普组红斑、鳞屑和厚度评分较模型组降低(均P<0.05)。ILC1和ILC3水平比较,模型组高于对照组,依那西普组低于模型组(均P<0.05); ILC2水平比较,模型组高于对照组,依那西普组低于模型组(均P<0.05),表明依那西普组可调控模型大鼠ILC亚群的表达。模型组T-bet、GATA3、RORγt、Caspase-3蛋白表达较对照组升高(均P<0.05),而Caspase-14、丝聚蛋白和外皮蛋白的蛋白表达降低(均P<0.05); 依那西普组T-bet、GATA3、RORγt、Caspase-3蛋白表达较模型组降低(均P<0.05),而Caspase-14、丝聚蛋白和外皮蛋白的蛋白表达升高(均P<0.05)。模型组IFN-γ、IL-5、IL-13、IL-17和IL-22 mRNA表达较对照组升高(均P<0.05),依那西普组IFN-γ、IL-5、IL-13、IL-17和IL-22 mRNA表达较模型组降低(均P<0.05)。结论:生物制剂依那西普可通过调控ILC亚群分型进而调控相应的转录因子和炎症因子的表达,从而达到减缓银屑病的作用。
Abstract:
Objective:To explore the effect of etanercept on the typing of innate lymphocyte subsets in a rat model of psoriasis.Methods:Thirty 6-week-old male Lewis rats were divided into control group,model group and etanercept group,10 rats in each group.The rat model of psoriasis was induced by imiquimod.The back phenotype of the rats was scored by the psoriasis skin lesion severity index.The levels of ILC1,ILC2 and ILC3 in the peripheral blood of the rats were analyzed by flow cytometry.The protein expressions of transcription factors T-bet,GATA3,RORγt,Caspase-3,Caspase-14,filaggrin and involucrin were detected by Western blot.The mRNA expressions of inflammatory cytokines in psoriatic skin tissues of rats were detected by RT-PCR.Results:The scores of erythema,scale and thickness in the model group were higher than those in the control group,and the scores of erythema,scale and thickness in the etanercept group were lower than those in the model group(all P<0.05).The levels of ILC1 and ILC3 in the model group were higher than those in the control group,and the levels of ILC1 and ILC3 in the etanercept group were lower than those in the model group(all P<0.05).The level of ILC2 in the model group was lower than that in the control group,and the level of ILC2 in the etanercept group was higher than that in the model group(all P<0.05).These results indicated that the etanercept group could regulate the expression of ILC subsets in model rats.The protein expressions of T-bet,GATA3,RORγt and Caspase-3 in the model group were higher than those in the control group,while the protein expressions of Caspase-14,filaggrin and involucrin were lower(all P<0.05).The protein expressions of T-bet,GATA3,RORγt and Caspase-3 in the etanercept group were lower than those in the model group,while the protein expressions of Caspase-14,filaggrin and involucrin were higher(all P<0.05).The mRNA expressions of IFN-γ,IL-5,IL-13,IL-17 and IL-22 in the model group were higher than those in the control group,and the mRNA expressions of IFN-γ,IL-5,IL-13,IL-17 and IL-22 in the etanercept group were lower than those in the model group(all P<0.05).Conclusion:The biological preparation etanercept can slow down psoriasis by regulating ILC subsets typing and then regulating the expression of corresponding transcription factors and inflammatory factors.
