摘要:
目的:探讨奥扎格雷钠对癫痫小鼠应激、神经损伤程度和神经功能重建相关因子及凝血指标的影响。方法:将60只小鼠随机分为三组:正常组,模型组(盐酸匹罗卡品180 mg/kg)、奥扎格雷钠组(20 mg/kg)。模型组及奥扎格雷钠组采用匹罗卡品使小鼠癫痫,采用酶联免疫吸附试验测定各组应激指标[去甲肾上腺素(NE)、皮质醇(Cor)]、神经损伤程度[神经元特异性烯醇化酶(NSE)、星形胶质源性蛋白(S100B)、泛素羧基末端水解酶 L1(UCH-L1)、胶质纤维酸性蛋白(GFAP)指标]、神经功能重建[脑源性神经营养因子(BDNF)、神经生长因子(NGF)],采用全自动凝血仪检测各项凝血功能[活化部分凝血酶原时间(APTT)、凝血酶原时间(PT)、凝血酶时间(TT)及纤维蛋白原(FIB)指标]。结果:与正常组相比,模型组NE、Cor表达水平明显升高(均P<0.05); 与模型组相比,奥扎格雷钠组NE、Cor表达水平明显降低(均P<0.05); 与正常组相比,模型组NSE、S100B、UCH-L1及GFAP水平明显升高(均P<0.05); 与模型组相比,奥扎格雷钠组NSE、S100B、UCH-L1、GFAP水平明显降低(均P<0.05); 与正常组相比,模型组BDNF、NGF水平明显降低(均P<0.05); 与模型组相比,奥扎格雷钠组神经功能重建的细胞因子BDNF、NGF的含量明显升高(均P<0.05); 与正常组相比,模型组凝血指标PT、TT、APTT明显降低,FIB明显升高,呈现相对高凝状态(均P<0.05); 与模型组相比,奥扎格雷钠组凝血指标PT、TT、APTT明显升高,FIB明显降低,呈现相对低凝状态(均P<0.05)。结论:奥扎格雷钠可有效减轻癫痫小鼠应激反应、缓解神经损伤症状,加强神经功能重建,改善凝血指标。
Abstract:
Objective:To explore the effects of ozagrel sodium on stress,nerve injury degree,nerve function reconstruction related factors and coagulation indexes in epileptic mice.Methods:A total of 60 mice were included in this study,and they were divided into normal group,model group(pirocarpine hydrochloride 180 mg/kg),and ozagrel sodium group(20 mg/kg).The model group and the ozagrel sodium group induced epilepsy by using pilocarpine.Enzyme-linked immunosorbent assay was used to determine the stress indexes [norepinephrine(NE),cortisol(Cor)],the nerve injury degree indexes [neuron-specific enolase(NSE),astrocyte-derived protein(S100B),ubiquitin carboxy-terminal hydrolase L1(UCH-L1),glial fibrillary acidic protein(GFAP)],nerve function reconstruction indexes [brain-derived neurotrophic factor(BDNF),nerve growth factor(NGF)].All coagulation functions indexes [activated partial prothrombin time(APTT),prothrombin time(PT),thrombin time(TT)and fibrinogen(FIB)] were measured by automatic coagulation apparatus.Results:Compared with the normal group,the expression levels of NE and Cor in the model group were significantly increased(all P<0.05).Compared with the model group,the expression levels of NE and Cor in the ozagrel sodium group were significantly decreased(all P<0.05).Compared with the normal group,NSE,S100B,UCH-L1 and GFAP levels in model group were significantly increased(all P<0.05).Compared with the model group,NSE,S100B,UCH-L1 and GFAP levels in ozagrel sodium group were significantly decreased(all P<0.05).Compared with the normal group,the levels of BDNF and NGF in the model group were significantly decreased(all P<0.05).Compared with the model group,the levels of cytokines BDNF and NGF in the ozagrel sodium group were significantly increased(all P<0.05).Compared with the normal group,the coagulation indexes PT,TT and APTT in the model group were significantly lower,and FIB was significantly higher,showing a relatively hypercoagulable state(all P<0.05).Compared with the model group,the coagulation indexes PT,TT and APTT were significantly higher,and FIB was significantly lower,showing a relatively low coagulation state(all P<0.05).Conclusion:Ozagrel sodium can effectively alleviate the stress response of epileptic mice,relieve the symptoms of nerve damage,strengthen the reconstruction of nerve function,and improve coagulation indexes.
