摘要:
目的:探讨参芍口服液对创伤性脑损伤后小鼠神经炎性反应的影响及作用机制。方法:54只C57小鼠随机等分为假手术组(Sham组)、创伤性脑损伤组(TBI组)和参芍口服液组(SS组),采用限制性皮层损伤的方法对TBI组和SS组小鼠进行损伤手术,造模6 h后,SS组给予5 ml参芍口服液灌胃干预,Sham组和TBI组用等量0.9%氯化钠溶液灌胃,持续7 d。采用mNSS评分对各组小鼠的神经功能进行评估,TUNEL法检测细胞的凋亡情况,酶联免疫吸附实验测定脑组织中炎症因子肿瘤坏死因子-α(TNF-α)、白介素1β(IL-1β)、IL-18的含量,用蛋白印迹法检测脑组织内NRLP3、Caspase-1的表达水平。结果:伤后1、3、7 d,对比Sham组,TBI组的mNSS评分明显升高(均P<0.05),皮层损伤区神经元凋亡率明显增加(均P<0.05); SS组的mNSS评分明显低于TBI组(P<0.05),皮层损伤区神经元凋亡率也较TBI组明显减少(P<0.05); 相比Sham组,伤后7 d,TBI组的炎症因子(IL-18、TNF-α、IL-1β)的表达均上升(均P<0.05),而SS组的炎症因子的表达量较TBI组明显下降(均P<0.05); 蛋白印迹法检测小鼠造模7 d后脑组织中NLRP3、Caspase-1含量,结果显示TBI组的NLRP3、Caspase-1含量明显高于Sham组,SS组的NLRP3、Caspase-1含量明显低于TBI组。结论:参芍口服液对小鼠的创伤性脑损伤有治疗作用,能够改善神经功能,减少神经元死亡比例,通过抑制NLRP3炎症小体的表达实现减轻炎性反应的效果。
Abstract:
Objective:To investigate the effect and mechanism of Shenshao oral liquid on neuroinflammatory response of mice after traumatic brain injury.Methods: Fifty-four rats were randomly divided into sham group(Sham group),traumatic brain injury group(TBI group)and Shenshao oral liquid group(SS group).Model of controlled cortical impact was used to duplicate the model of traumatic brain injury.SS group was given 5 ml Shenshao oral liquid by gavage 6 hours after operation.Six hours after modeling,SS group received 5 ml Shenshao oral liquid by gavage,while Sham group and TBI group received the same amount of 0.9% sodium chloride solution for 7 days.The neural function was evaluated by mNSS score.Apoptosis was detected by TUNEL method.The contents of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and IL-18 in brain tissue were determined by enzyme-linked immunosorbent assay.The expression levels of NRLP3 and Caspase-1 in brain tissue were detected by Western blot.Results:On the 1st,3rd and 7th day after injury,compared with Sham group,the mNSS score and the proportion of TUNEL positive cells of TBI group were significantly increased(all P<0.05); the mNSS score and the proportion of TUNEL positive cells of SS group were significantly lower than those of TBI group(all P<0.05).At 7 days after injury,the expression of inflammatory factors(IL-18,TNF-α,IL-1β)in TBI group increased compared with Sham group,while the expression of inflammatory factors in SS group decreased significantly compared with TBI group(all P<0.05); NLRP3 and Caspase-1 contents in TBI group were significantly higher than those in Sham group,while NLRP3 and Caspase-1 contents in SS group were significantly lower than those in TBI group(all P<0.05).Conclusion:Shenshao oral liquid has therapeutic effect on traumatic brain injury in mice.It can improve neurological function,reduce the number of neuronal death,and reduce the inflammatory reaction by inhibiting the expression of NLRP3 inflammasome.
