摘要:
目的:探讨RNA结合蛋白震颤同系物-5(QKI-5)在卵巢癌细胞中的表达以及对卵巢癌细胞增殖和凋亡的影响。方法:通过免疫印迹实验(Western blot)检测正常卵巢上皮细胞和卵巢癌细胞中QKI-5的表达差异,并选取QKI-5相对表达水平最低的卵巢癌细胞进行过表达慢病毒感染,构建稳定转染细胞系后通过细胞增殖和毒性分析(CCK8)和平板克隆实验检测细胞增殖和克隆能力的变化,通过流式细胞仪检测过表达QKI-5对卵巢癌细胞凋亡的影响。结果:Western blot实验结果显示,在卵巢癌细胞中QKI-5的表达水平相比正常卵巢上皮细胞显著降低,三株卵巢癌细胞中A2780细胞的QKI-5表达水平相对最低,通过慢病毒感染成功构建了稳定过表达QKI-5的A2780细胞系。CCK8和平板克隆实验结果显示,过表达QKI-5基因后A2780细胞增殖活力和克隆形成能力受到抑制(P<0.05)。流式细胞仪检测细胞凋亡率结果显示,QKI-5表达水平的增加能够显著促进A2780细胞的凋亡(P<0.05)。结论:RNA结合蛋白QKI-5在卵巢细胞中表达水平异常降低,QKI-5表达特异性扩增后能够抑制卵巢癌细胞A2780增殖和克隆形成能力并促进癌细胞发生凋亡。
Abstract:
Objective:To investigate the expression of RNA binding protein quaking homologue-5(QKI-5)in ovarian cancer cells and its effect on proliferation and apoptosis of ovarian cancer cells.Methods:The expression of QKI-5 in normal ovarian epithelial cells and ovarian cancer cells was detected by Western blot,and the ovarian cancer cells with the lowest relative expression level of QKI-5 were selected for lentivirus infection.After stable transfected cell lines were constructed,cell proliferation and cloning ability were detected by CCK8 and plate cloning experiments.The effect of QKI-5 overexpression on apoptosis of ovarian cancer cells was detected by flow cytometry.Results:Western blot showed that the expression level of QKI-5 in ovarian cancer cells was significantly lower than that in normal ovarian epithelial cells.The expression level of QKI-5 in A2780 cells was the lowest among the three ovarian cancer cells.The A2780 cell line with stable overexpression of QKI-5 was successfully constructed by lentivirus infection.CCK8 and plate cloning experiments showed that the proliferation activity and clone formation ability of A2780 cells were inhibited after QKI-5 gene overexpression(all P<0.05).The apoptosis rate of A2780 cells was detected by flow cytometry,and the increase of QKI-5 expression level significantly promoted the apoptosis of A2780 cells(P<0.05).Conclusion:The expression level of RNA binding protein QKI-5 is abnormally decreased in ovarian cells.The specific amplification of QKI-5 expression can inhibit the proliferation and clone formation,and promote apoptosis of ovarian cancer cell A2780.
