摘要:
目的:研究胃泌素释放肽(GRP)及其受体(GRPR)在兔耳增生性瘢痕组织中的表达情况,并探讨其表达与瘢痕组织内巨噬细胞浸润的相关性。方法:18只新西兰大鼠随机分为对照组、瘢痕A组和瘢痕B组,瘢痕A组在每个兔耳建立直径0.6 cm创面1个,瘢痕B组在每个兔耳建立直径1.0 cm的创面1个,对照组兔耳不建立创面。分别在创面建立1个月(增生期)、2个月(消退期)和3个月(成熟期),每组取3只兔耳瘢痕组织检测GRP、GRPR、巨噬细胞表面标志物(F4/80)以及巨噬细胞浸润数目。结果:GRP、GRPR、F4/80 mRNA表达水平和巨噬细胞浸润数目在增生期、消退期和成熟期兔耳增生性瘢痕组织均显著高于对照组(均P<0.05),且其在瘢痕A组中均低于瘢痕B组(P<0.05)。GRP、GRPR、F4/80 mRNA表达水平和巨噬细胞浸润数目在兔耳瘢痕组织中随时间延长而降低。在兔增生性瘢痕组织内,GRP和GRPR mRNA表达水平与F4/80 mRNA表达水平、巨噬细胞浸润数目均呈正相关(P<0.05)。结论:GRP和GRPR在兔耳增生性瘢痕组织内表达升高,并且其表达升高与创面大小有关,均随创面愈合而降低,且与增生性瘢痕组织内巨噬细胞浸润呈正相关。
Abstract:
Objective:To study the expression of gastrin-releasing peptide(GRP)and its receptor(GRPR)in hypertrophic scar tissue of rabbit ears,and to explore its correlation with macrophage infiltration in scar tissue.Methods: Eighteen New Zealand rats were randomly divided into control group,scar group A and scar group B.In scar group A,one wound with a diameter of 0.6 cm was established in each rabbit ear.One wound with a diameter of 1.0 cm was established in each rabbit ear of scar group B.The rabbit ears of the control group did not establish wounds.At 1 month(proliferation phase),2 months(involution phase)and 3 months(mature phase)of wound establishment,three rabbit ear scar tissues were taken from each group to detect GRP,GRPR,macrophage surface markers(F4/80)and the number of macrophage infiltration.Results:The expression levels of GRP,GRPR,F4/80 mRNA and the number of infiltrated macrophages were significantly higher in hypertrophic scar tissue of rabbit ears in the proliferative,regression and mature stages than those in the control group,and they were significantly higher in scar group A than those in the control group(all P<0.05).The expression levels of GRP,GRPR,F4/80 mRNA and the number of macrophages infiltrated in rabbit ear scar tissue decreased with time.In rabbit hypertrophic scar tissue,the expression levels of GRP and GRPR mRNA were positively correlated with the expression level of F4/80 mRNA and the number of infiltrated macrophages(all P<0.05).Conclusion:The expressions of GRP and GRPR were increased in hypertrophic scar tissue of rabbit ears,and the increased expression was related to the size of the wound,decreased with wound healing,and was positively correlated with the infiltration of macrophages in the hypertrophic scar tissue.
