摘要:
目的:评价丙酮酸乙酯(EP)对脑缺血再灌注损伤时小胶质细胞M1/M2极化表型的影响。方法:雄性SD大鼠72只,体重220~250 g,采用随机数字法分为三组:假手术组(Sham组)、大脑中动脉栓塞组(MCAO组)和丙酮酸乙酯+大脑中动脉栓塞组(EP+MCAO组)。MCAO组和EP+MCAO组采用Zea-Longa法制备大鼠MCAO模型,并在栓塞大脑中动脉90 min后拔除线栓,分别静脉注射0.9%氯化钠溶液或EP溶液。Sham组与其他两组在同一时间静脉注射0.9%氯化钠溶液。分别于造模后24、48、72 h时,进行神经功能缺损评分(NSS); 处死大鼠进行TTC染色检测脑梗死容积; 取缺血侧脑组织匀浆,采用酶联免疫法(ELISA)检测炎症因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)及脑源性神经营养因子(BDNF)的浓度; 采用免疫荧光染色法和Western blot法检测小胶质细胞M1型极化标志物诱导型一氧化氮合酶(iNOS)和M2型极化标志物精氨酸酶-1(Arg-1)的表达。结果:与Sham组比较,MCAO组和EP+MCAO组NSS、脑梗死容积、IL-1β、IL-6、IL-10、BDNF浓度升高、iNOS和Arg-1表达上调(均P<0.05); 与MCAO组比较,EP+MCAO组NSS、脑梗死容积、IL-1β、IL-6浓度降低,iNOS表达下调(均P<0.05),IL-10、BDNF浓度升高,Arg-1表达上调(均P<0.05)。结论:EP减轻脑缺血再灌注损伤的机制可能与调控小胶质细胞极化,抑制炎性反应有关。
Abstract:
Objective:To evaluate the effects of ethyl pyruvate on microglial polarization after ischemia/reperfusion brain injury in rats.Methods: Seven-two male Sprague-Dawley rats,weighing 220 to 250 g,were divided into three groups using a random number table method:sham operation group(Sham group),middle cerebral artery occlusion group(MCAO group),and ethyl pyruvate plus MCAO group(EP+MCAO group).MCAO models were established by using Zea-Longa method in MCAO and EP+MCAO group,after suture removed,normal saline or ethyl pyruvate solution was injected respectively in MCAO and EP+MCAO group.At 24,48 and 72 hours after establishing the model,the neurological function was evaluated with the neurologic severity score(NSS).The rats were then sacrificed,and the cerebral infarction volume was evaluated by TTC staining.The homogenates of ischemic brain tissue were collected and the concentrations of inflammatory cytokines interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-10(IL-10)and brain-derived neurotrophic factor(BDNF)were detected by enzyme-linked immunosorbent assay(ELISA).The expression of M1 polarization marker inducible nitric oxide synthase(iNOS)and M2 polarization marker arginase -1(Arg-1)in microglia was determined by immunofluorescence staining and Western blot.Results:Compared with Sham group,the concentrations of NSS,cerebral infarction volume,IL-1β,IL-6,IL-10,BDNF and iNOS were increased in MCAO group and EP+MCAO group(all P<0.05).Compared with MCAO group,the concentrations of NSS,cerebral infarction volume,IL-1β,IL-6 and iNOS were decreased in EP+MCAO group(all P<0.05),and Arg-1 expression was up-regulated(all P<0.05).Conclusion:The mechanism of ethyl pyruvate in reducing cerebral ischemia-reperfusion injury may be related to regulating microglia polarization and inhibiting inflammatory reaction.
