摘要:
目的:探讨细胞周期蛋白依赖性激酶亚基(CKS)2通过干预线粒体功能促进宫颈癌细胞增殖的机制。方法:对数生长期的宫颈癌SiHa细胞分为三组:空白对照组(不进行转染)、阴性对照组(转染Hs-NC siRNA)与CKS2 siRNA组(转染Hs-CKS2 siRNA),采用MTT法检测细胞增殖活性,Transwell检测细胞侵袭及转移,线粒体膜电位检测试剂盒检测线粒体膜电位,qPCR检测CKS2与线粒体整合蛋白2(Mfn2)mRNA水平,Western blot法检测Mfn2、CKS2蛋白水平。结果:转染后24、36 h,CKS2 siRNA组的细胞增殖指数、细胞侵袭及转移指数低于空白对照组、阴性对照组(均P<0.05); 转染后24、36 h,CKS2 siRNA组的细胞线粒体膜电位水平高于空白对照组、阴性对照组(均P<0.05); 转染后24、36 h,CKS2 siRNA组的CKS2与Mfn2 mRNA、蛋白相对表达水平低于空白对照组、阴性对照组(均P<0.05)。结论:抑制CKS2的表达可通过干预线粒体膜电位水平,抑制宫颈癌细胞Mfn2的表达,从而抑制宫颈癌细胞增殖、侵袭与转移。
Abstract:
Objective:To investigate the mechanism of cyclin-dependent kinase subunit(CKS)2 promoting the proliferation of cervical cancer cells by interfering with mitochondrial function.Methods:Cervical cancer SiHa cells in logarithmic growth phase were divided into three groups:blank control group(without transfection),negative control group(transfected with Hs-NC siRNA)and CKS2 siRNA group(transfected with Hs-CKS2 siRNA).Cell proliferation activity were detected by MTT method,and cell invasion and metastasis were detected by Transwell,and mitochondrial membrane potential was detected by mitochondrial membrane potential detection kit,and CKS2 and Mfn2 mRNA levels were detected by qPCR,and Mfn2 and CKS2 protein levels were detected by Western blot.Results:At 24 and 36 hours after transfection,the cell proliferation index,cell invasion and metastasis index of the CKS2 siRNA group were lower than those of the blank control group and the negative control group,and the cell mitochondrial membrane potential level of the CKS2 siRNA group were higher than that of the blank control group and the negative control group,and the relative expression levels of CKS2 and Mfn2 mRNAs and proteins in the CKS2 siRNA group were lower than those of the blank control group and the negative control group(all P<0.05).Conclusion:Inhibiting the expression of CKS2 can inhibit the expression of cervical cancer cells Mfn2 by interfering with the level of mitochondrial membrane potential,thereby inhibiting the proliferation,invasion and metastasis of cervical cancer cells.
