摘要:
目的:研究维生素K2对去卵巢股骨干骨折模型大鼠骨折愈合及骨密度的影响。方法:将90只雌性SD大鼠随机分为假手术组、骨折组及干预组,每组30只。三组大鼠均实施卵巢切除干预,其中骨折组与干预组均建立股骨干骨折模型。干预组采用维生素K2灌胃,假手术组与骨折组均予以等量0.9%氯化钠溶液,连续干预5周。比较三组大鼠骨折愈合状况、骨密度[包括全长骨密度(tBMD)、中1/3段骨密度(mBMD)以及远1/3段骨密度(dBMD)],以及骨代谢指标[包括骨钙素(BGLAP)、碱性磷酸酶(ALP)及抗酒石醋酸酸性磷酸酶(TRACP)]、血清血管内皮生长因子(VEGF)和骨形成蛋白-2(BMP-2)表达水平。结果:X线检查发现,骨折组大鼠不完全愈合,而干预组存在明显的骨痂组织形成、重塑以及骨愈合。骨折组大鼠右股骨tBMD、mBMD及dBMD均低于假手术组,而干预组各项指标水平均高于骨折组(均P<0.05)。骨折组大鼠BGLAP、ALP及抗TRACP水平均明显高于骨折组,而干预组大鼠各项指标水平均明显低于骨折组(均P<0.05)。骨折组大鼠血清VEGF、BMP-2表达水平均明显低于假手术组,而干预组VEGF、BMP-2表达水平均高于骨折组(均P<0.05)。结论:维生素K2可促进去卵巢股骨干骨折模型大鼠的骨折愈合并增加骨密度。
Abstract:
Objective:To study the effect of vitamin K2 on the fracture healing and bone density in ovariectomized femoral shaft fracture model rats.Methods:90 female SD rats were randomly divided into sham operation group,fracture group and intervention group,with 30 rats in each group.Ovariectomies were performed in all three groups,and femoral shaft fracture models were established in the fracture group and the intervention group.The intervention group was given intragastric administration of vitamin K2,and the sham operation group and the fracture group were given the same amount of 0.9% sodium chloride solution,and all the rats were treated for 5 weeks.The fracture healing status,bone density(including tBMD,mBMD and dBMD),and the expression levels of bone metabolism indexes(including BGLAP,ALP and TRACP),serum VEGF and BMP-2 in the three groups of rats were compared.Results:X-ray examination showed that the fracture group rats were not completely healed,and the intervention group had obvious callus formation,remodeling and bone healing.The tBMD,mBMD and dBMD of right femur in the fracture group were lower than those in the sham operation group,and the levels of the above indicators in the intervention group were higher than those in the fracture group(all P<0.05).The BGLAP,ALP and TRACP levels in the fracture group were significantly higher than those in the fracture group,and the levels of the above indexes in the intervention group were significantly lower than those in the fracture group(all P<0.05).Serum VEGF and BMP-2 expression levels in the fracture group were significantly lower than those in the sham operation group,and the expression levels of VEGF and BMP-2 in the intervention group were higher than those in the fracture group(all P<0.05).Conclusion:Vitamin K2 can promote fracture healing and increase bone density in ovariectomized femoral shaft fracture model rats.
