摘要:
目的:探讨尼古丁是否通过α7亚型烟碱型乙酰胆碱受体(α7 nAChR)影响人牙周膜干细胞(PDLSCs)成骨分化潜能。方法:极限稀释法获得PDLSCs。茜素红染色评估细胞分化形成矿化结节的能力。采用实时定量PCR(qRT-PCR)和Western blot检测成骨相关基因ALP、OCN、BSP、Runx2及其蛋白的表达情况。采用Western blot检测α7 nAChR及其下游通路相关蛋白表达情况。结果:尼古丁以浓度依赖的方式影响PDLSCs成骨分化。尼古丁通过上调α7 nAChR表达进一步激活Wnt/β-catenin信号通路,从而导致PDLSCs成骨分化能力降低。抑制α7 nAChR及阻断Wnt通路能够逆转尼古丁对细胞分化的负性调控作用。结论:尼古丁可通过α7 nAChR抑制PDLSCs成骨分化,这可能是吸烟相关性牙周炎的发病机制之一。
Abstract:
Objective:To investigate whether nicotine affected the osteogenic differentiation potential of human periodontal ligament stem cells(PDLSCs)through the α7 nicotinic acetylcholine receptor(α7 nAChR).Methods:PDLSCs were harvested by limiting dilution technique.The ability of mineralized nodule formation was assessed by alizarin red staining.Real-time quantitative PCR(qRT-PCR)and Western blot were used to detect the expression of ostegenic related genes ALP,OCN,BSP,Runx2 and their proteins.Western blot was used to detect the expression of α7 nAChR and related proteins in downstream pathways.Results:Nicotine deteriorated the ostegenic differentiation of PDLSCs in a concentration-dependent manner.Nicotine further activated the Wnt/β-catenin signaling pathway by up-regulating the expression of α7 nAChR,which led to a decrease in the osteogenic differentiation ability of PDLSCs.Inhibiting α7 nAChR and blocking the Wnt pathway could reverse the negative regulatory effect of nicotine on cell differentiation.Conclusion:Nicotine can inhibit the osteogenic differentiation of PDLSCs through α7 nAChR,which may be one of the pathogenesis of smoking-related periodontitis.
