摘要:
目的:探讨干眼症小鼠泪液中胸腺基质淋巴细胞生成素(TSLP)和白介素18(IL-18)的水平变化及相关机制。方法:64只健康雄性BALB/c小鼠随机分为对照组与模型组各32只。模型组小鼠双眼各滴入5 μl 0.2%苯扎氯铵溶液,每天2次,连续15 d,诱导小鼠干眼症模型,对照组以等剂量的磷酸盐缓冲液滴眼。采用酚红棉线检测泪液分泌量,行角膜荧光素染色和HE染色,记录泪膜破裂时间,采用酶联免疫法检测泪液中IL-18、TSLP水平。结果:两组实验第1天的泪液分泌实验(SIT)、泪膜破裂时间(BUT)、角膜荧光素染色(FL)评分、IL-18、TSLP水平对比差异无统计学意义(均P>0.05),模型组实验第15天的SIT与BUT值低于对照组,也低于实验第1天,对比差异均有统计学意义(均P<0.05); 模型组实验第15天的FL评分、泪液IL-18、TSLP水平均显著高于对照组和实验第1天(均P<0.05)。结论:TSLP在干眼症小鼠泪液中呈现高表达水平,可能抑制神经递质的释放,促进泪液IL-18的表达,降低泪液分泌,从而诱发干眼症的发生。
Abstract:
Objective:To investigate the changes and related mechanisms of thymic stromal lymphopoietin(TSLP)and interleukin-18(IL-18)in tears of mice with dry eye.Methods:64 cases of healthy male BALB/c mice were randomly divided into control group and model group,with 32 cases in each.Mice in the model group were injected into both eyes with 5 μl 0.2% benzalkonium chloride solution,twice a day for 15 days,to induce the mouse dry eye model.The control group was dripped with the same dose of phosphate buffer.The tear secretion was detected by phenol red cotton thread,corneal fluorescein staining and HE staining were performed,the tear film rupture time was recorded,and the levels of IL-18 and TSLP in tear were detected by enzyme-linked immunosorbent assay.Results:There were no significant differences in SIT,BUT,FL score,tear IL-18,TSLP levels on the 1st day of the experiment between the two groups(all P>0.05).The SIT and BUT values on the 15th day of the experiment in the model group were lower than those of the control group,and also less than those on the 1st day of the experiment(all P<0.05).The FL score,IL-18 and TSLP levels of the model group on the 15th day of the experiment were higher than those of the control group,and also higher than those on the 1st day of the experiment(all P<0.05).Conclusion:TSLP is highly expressed in the tears of mice with dry eye,which may inhibit the release of neurotransmitters,promote the expression of IL-18 in tears,and reduce secretion of tears,so as to induce the occurrence of dry eye.
