摘要:
目的:探讨阿霉素诱导心脏损伤模型大鼠的超声心功能指标与核因子κB(NF-κB)p65表达水平的关系。方法:清洁级雄性健康成年Sprague-Dawley(SD)大鼠72只随机平分为三组:对照组、模型A组、模型B组,三组分别给予腹腔注射0、1、5 ml/kg阿霉素,在造模第1周与第2周每组各处死12只大鼠,检测大鼠的超声心功能指标与NF-κB p65表达水平。结果:模型A组、模型B组造模第1周与第2周的左室收缩压(LVSP)、左室舒张末压(LVEDP)、测丙二醛(MDA)高于对照组(均P<0.05),左室射血分数(LVEF)、左室短轴缩短率(LVFS)、超氧化物歧化酶(SOD)、NF-κB p65低于对照组(均P<0.05),模型A组与模型B组比较差异均有统计学意义(P<0.05)。在模型B组中,造模第2周的NF-κB p65蛋白相对表达水平与LVSP、LVEDP、LVEF、LVFS都存在相关性(均P<0.05)。结论:阿霉素诱导心脏损伤模型大鼠多伴随有超声心功能指标的异常与NF-κB p65表达水平下降,两者存在相关性。
Abstract:
Objective:To investigate the relationship between the echocardial function indexes and the expression of nuclear factor kappa-B(NF-κB)p65 in model rats with heart injury induced by adriamycin.Methods:72 clean-grade male healthy adult Sprague-Dawley(SD)rats were randomly divided into control group,model 1 group,and model 2 group.The three groups were given intraperitoneal injections of 0,1and 5 ml/ kg of adriamycin respectively.There were 12 rats in each group died at the 1st and 2nd week of modeling,and the echocardiographic function indexes and NF-κB p65 expression level of the model rats were detected.Results:The left ventricular systolic blood pressure(LVSP),left ventricular end diastolic pressure(LVEDP),and malondialdehyde(MDA)at the 1st and 2nd week of modeling in the model 1 group and model 2 group were higher than those in the control group(all P<0.05),left ventricular ejection fraction(LVEF),left ventricular short axis shortening rate(LVFS),superoxide dismutase(SOD)and NF-κB p65 were lower than those of the control group(all P<0.05),the difference between the model 1 group and model 2 group were also statistically significant(all P<0.05).In the model 2 group,the relative expression level of NF-κB p65 protein at the 2nd week of modeling were correlated with LVSP,LVEDP,LVEF and LVFS(all P<0.05).Conclusion:Adriamycin-induced heart injury model rats are mostly accompanied by abnormal echocardiographic indexes and decreased expression of NF-κB p65,and there are correlation between NF-κB p65 and echocardiographic indexes.
