摘要:
目的:采用外周血血小板裂解产物(PB-PL)替代胎牛血清(FBS),观察其对人脐带间充质干细胞(MSCs)增殖、分化的影响。方法:采用PB-PL(PB-PL组)和FBS(FBS组)分别培养扩增人脐带MSCs。采用台盼蓝染色法检测两组MSCs细胞活性。比较两组MSCs细胞倍增时间。将两组MSCs分别置于成骨、成软骨和成脂定向诱导培养体系中,向骨、软骨和脂肪方向分化。采用茜素红染色检测成骨分化能力,采用阿新兰染色检测成软骨分化能力,采用油红O染色检测细胞成脂情况。比较两组MSCs集落生成能力。结果:采用含FBS或PB-PL的培养基能够分离和培养脐带MSCs。两组MSCs形态能维持8代以上。PB-PL组MSCs显示出较高的增殖率。采用FBS和PB-PL扩增的MSCs均表现出向骨、软骨和脂肪方向分化能力。结论:PB-PL作为培养和扩增人脐带MSCs的添加剂展现出良好的保持细胞原始属性、支持细胞体外增殖和分化的能力,可以作为非异种培养基添加剂替代FBS用于体外扩增人脐带MSCs。
Abstract:
Objective:To replace FBS with peripheral blood platelet lysate(PB-PL),and to observe its effect on the proliferation and differentiation of human umbilical cord mesenchymal stem cells(MSCs).Methods:PB-PL(PB-PL group)and FBS(FBS group)were used to culture and expand human umbilical cord MSCs.The cell viability of the two groups of MSCs was detected by trypan blue staining.The doubling time of MSCs between the two groups was compared.The two groups of MSCs were placed in a directional induction culture system for osteogenesis,cartilage and adipogenesis,and differentiated into bone,cartilage and fat.Alizarin red staining was used to detect osteogenic differentiation ability,azulam staining was used to detect cartilage differentiation ability,and oil red O staining was used to detect cell adipogenesis.The two groups of MSCs colony forming ability was compared.Results:The umbilical cord MSCs could be isolated and cultured by using medium containing FBS or PB-PL,and the morphology of the two groups of MSCs could maintain more than 8 generations.MSCs in the PB-PL group showed a higher proliferation rate.MSCs amplified with FBS and PB-PL showed trilinear differentiation ability. Conclusion:PB-PL,as an additive for the cultivation and amplification of human umbilical cord MSCs,showed a good ability to maintain the original cell properties and support the proliferation and differentiation of cells in vitro.It can be used as a non-heterogeneous medium additive to replace FBS for the amplification of human umbilical cord MSCs in vitro.
