摘要:
目的:观察大黄、赤芍注射液对急性肝衰竭(ALF)模型大鼠肝功能及肝再生指标的影响,并探讨相关机制。方法:构建ALF大鼠模型后,选取100只随机分为五组,每组20只。比较造模后24、48 h各组大鼠血清肝功能指标[天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)及总胆红素(TBIL)]及肝再生指标[有丝分裂指数(MI)、增殖指数(PI)和增殖细胞核抗原(PCNA)]。 结果:造模后,对照组大鼠精神萎靡、行动迟缓,药物干预组大鼠总体表现较对照组好。造模后24、48 h,对照组ALT、AST、TBIL水平高于其他四组; 对照组MI和PI低于其他四组,生长素组与低剂量组高于中剂量组与高剂量组; 低剂量组、高剂量组和生长素组PCNA阳性细胞数高于对照组(均P<0.05)。干预药物均能够促进肝细胞活化和增殖,且以低剂量组与生长素组效果最佳。结论:大黄、赤芍注射液可改善ALF大鼠肝功能,提高大鼠存活率,其机制可能与提高大鼠肝细胞MI、PI、PCNA水平,促进肝细胞再生有关。
Abstract:
Objective:To observe the effect of Rhubarb and Radix paeoniae rubra injection on liver function and liver regeneration indexes in rats with acute liver failure,and to explore its mechanism.Methods:After the ALF rat model was constructed,100 rats were selected and randomly divided into five groups,with 20 rats in each group.The serum liver function indexes(AST,ALT and TBIL)and liver regeneration indexes(MI,PI and PCNA)at 24 and 48 hours after modeling were compared among five groups.Results:After the model was established,the rats in the control group were lethargic and slow to act.The overall performance of the rats in the drug intervention group was better than that in the control group.24 and 48 hours after modeling,the ALT,AST,and TBIL levels of the control group were higher than those of other four groups; the MI and PI of the control group were lower than those of other four groups,and the auxin group and the low-dose group were higher than the medium-dose group and the high-dose group; the number of PCNA-positive cells in the low-dose group,high-dose group and auxin group was higher than that in the control group(all P<0.05).Intervention drugs could promote liver cell activation and proliferation,and the low-dose group and the auxin group had the best effect.Conclusion:Rhubarb and Radix paeoniae rubra injection can improve the liver function of ALF rats and increase the survival rate of rats.The mechanism may be related to increasing the levels of MI,PI,PCNA in rat liver cells and promoting liver cell regeneration.
