摘要:
目的:观察微小RNA-215-5p(microRNA-215-5p,miR-215-5p)与真核细胞翻译起始因子2a(eIF2a)的靶向关系,探讨两者表达的临床意义。方法:选择骨肉瘤细胞系Saos2,构建阴性对照组、无关序列组、miR-215-5p mimic组、miR-215-5p inhibitor组,应用Western blot检测各组中eIF2a的表达。应用双荧光素酶报告基因实验检测miR-215-5p与eIF2a的结合情况。收集骨肉瘤术后标本38例,应用实时荧光定量PCR法(qPCR)检测miR-215-5p的表达,应用免疫组化法检测eIF2a的表达。结果:miR-215-5p mimic组中eIF2a的表达明显低于阴性对照组和无关序列组,miR-215-5p inhibitor组中eIF2a的表达明显高于阴性对照组和无关序列组。双荧光素酶报告基因实验显示miR-215-5p与eIF2a具有靶向关系。骨肉瘤中miR-215-5p的表达量、eIF2a的阳性率在不同肿瘤最大径和病变级别中的差异有统计学意义。相关分析显示miR-215-5p与eIF2a呈负相关性。结论:骨肉瘤细胞中miR-215-5p和eIF2a具有靶向关系,miR-215-5p与eIF2a的异常表达可能参与肿瘤的进展。
Abstract:
Objective:To observe the targeting relationship between microRNA-215-5p(miR-215-5p)and eukaryotic translation initiation factor 2a(eIF2a)in osteosarcoma cells,and to explore the clinical significance of their expression.Methods:Osteosarcoma cell line Saos2 was selected to construct negative control group,unrelated sequence group,miR-215-5p mimic group and miR-215-5p inhibitor group.The expression of eIF2a was detected by Western blot in each group.The dual luciferase reporter gene assay was used to detect the binding of miR-215-5p and eIF2a.38 postoperative specimens of osteosarcoma were collected,and the real-time fluorescent quantitative PCR(qPCR)was used to detect the expression of miR-215-5p,and the immunohistochemistry was used to detect the expression of eIF2a.Results:The expression of eIF2a in miR-215-5p mimic group was significantly lower than that in negative control group and unrelated sequence group,and the expression of eIF2a in miR-215-5p inhibitor group was significantly higher than that in negative control group and unrelated sequence group.Targeting relationship was found between miR-215-5p and eIF2a by double luciferase reporter gene assay.The expressions of miR-215-5p and eIF2a were significantly different in different tumor sizes and clinical stages of osteosarcoma.Negative correlation was found between miR-215-5p and eIF2a.Conclusion:Targeting relationship is found between miR-215-5p and eIF2a in osteosarcoma cells.The abnormal expression of miR-215-5p and eIF2a may be involved in the progression of osteosarcoma.
