摘要:
目的:探究小白菊内酯(PTL)对小儿血管瘤内皮细胞凋亡的影响。方法:MTS法筛选PTL促进小儿血管瘤内皮细胞凋亡的适宜浓度,细胞凋亡流式细胞法检测PTL对小儿血管瘤内皮细胞凋亡的影响,实时荧光定量 PCR(qPCR)和Western blot法检测PTL对小儿血管瘤内皮细胞凋亡相关蛋白表达的影响。结果:20、40 μmol/L PTL可显著降低小儿血管瘤内皮细胞活性(均P<0.01),20、40 μmol/L PTL可显著促进小儿血管瘤内皮细胞凋亡; 选择20 μmol/L PTL 用于后续实验,Tunel实验结果表明PTL显著促进小儿血管瘤内皮细胞凋亡(P<0.01),qPCR和Western blot表明PTL显著促进小儿血管瘤内皮细胞Bax和Cleaved-Caspase 3蛋白表达,抑制Bcl-2 mRNA和蛋白表达(均P<0.01)。结论:PTL通过抑制Bcl-2,促进Bax和Cleaved-Caspase 3等凋亡相关蛋白表达进而诱导小儿血管瘤内皮细胞凋亡。
Abstract:
Objective:To explore the effect of parthenolide(PTL)on the apoptosis of endothelial cells in children with hemangioma Methods:MTS method was used to screen the appropriate concentration of PTL to promote endothelial cells apoptosis in children with hemangioma.Flow cytometry was used to detect the effect of PTL on the apoptosis of endothelial cells in children with hemangioma.Real time PCR and Western blot methods were used to detect the effect of PTL on apoptosis-related protein of endothelial cells in children with hemangioma.Results:20 and 40 μmol/L PTL significantly reduced endothelial cell activity,and significantly promoted endothelial cell apoptosis in children with hemangioma(all P<0.01).20 μmol/L PTL was selected for follow-up experiments.Tunel experiment results showed that PTL significantly promoted endothelial cell apoptosis in children with hemangioma(P<0.01).Real time PCR and Western blot showed that PTL significantly promoted the expression of Bax and Cleaved-Caspase 3 proteins in endothelial cells in children with hemangioma,and inhibited the expression of Bcl-2 mRNA and protein(all P<0.01).Conclusion:PTL inhibited the expression of Bcl-2,to promote the expression of apoptosis-related proteins such as Bax and Cleaved-Caspase 3,and induced the apoptosis of endothelial cells in children with hemangioma.
