摘要:
目的:探讨构建急性肝衰竭(ALF)大鼠模型的最佳方式。方法:将30只SPF级SD雄性大鼠随机分为对照组、手术组及模型组,每组10只。对照组常规饲养,不予手术处理。手术组采用单纯60%肝切除术。模型组行60%肝切除术后予以20 mg/100 g D-氨基半乳糖(D-GalN)和10 μg/100 g脂多糖(LPS)腹腔注射。观察造模过程中各组大鼠死亡情况。比较各组大鼠造模后24、48 h血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)及总胆红素(TBIL)水平。通过HE染色观察各组大鼠造模后24、48 h肝组织病理学变化。结果:造模后48 h内,仅模型组大鼠非正常死亡3只,生存率为70%。造模后24、48 h,手术组与模型组大鼠ALT、AST、TBIL水平均高于对照组,模型组大鼠ALT、AST、TBIL水平均高于手术组(均P<0.05)。HE染色结果显示,模型组造模后24 h肝细胞呈灶状坏死,结构紊乱,可见肝细胞水肿; 造模后48 h肝细胞呈大块状坏死,结构紊乱,较对照组及手术组更符合肝衰竭模型要求。结论:60%肝切除术联合D-GalN及LPS腹腔注射操作简便,大鼠死亡率低,成模率高,为制作ALF大鼠模型的较理想方法。
Abstract:
Objective:To explore the best way to construct acute rat liver failure(ALF)model.Methods:30 SPF SD male rats were randomly divided into control group,operation group and model group,with 10 rats in each group.The normal control group was fed routinely without surgery.The rats in the operation group received 60% hepatectomy alone,and the rats in the model group received 20 mg/ 100 g D-GalN and 10 μg/100 g LPS intraperitoneal injection after 60% hepatectomy.The death of rats in each group was observed during modeling.The levels of serum ALT,AST and TBIL were compared at 24 and 48 hours after modeling.HE staining was used to observe the histopathological changes of the rats in each group at 24 and 48 hours after modeling.Results:Within 48 hours after modeling,only 3 rats in the model group died abnormally,and the survival rate was 70%.The levels of ALT,AST and TBIL in the operation group and model group were higher than those in the control group at 24 and 48 hours after modeling,and the levels of ALT,AST and TBIL in the model group were higher than those in the operation group(all P<0.05).He staining results showed that the hepatocytes in rats of the model group showed focal necrosis,structural disorder and hepatocyte edema at 24 hours after modeling,and massive necrosis,structural disorder at 48 hours after modeling,which was more consistent with the requirements of liver failure model than the normal group and the operation group.Conclusion:60% hepatectomy combined with D-GalN and LPS intraperitoneal injection has the advantages of simple operation,low mortality and high modeling rate,which is an ideal method for the establishment of rat model of ALF.
