摘要:
目的:探索环状RNA hsa_circ_103809在骨肉瘤中的表达及生物学作用。方法:采用荧光定量PCR(qRT-PCR)检测骨肉瘤组织及细胞系中hsa_circ_103809表达水平。将骨肉瘤细胞分为两组,实验组转染hsa_circ_103809 siRNA序列,对照组转染siRNA阴性对照(NC)序列。采用CCK-8增殖实验和Transwell侵袭与迁移实验检测两组细胞的增殖、侵袭和迁移能力。分析骨肉瘤组织中hsa_circ_103809表达水平与骨肉瘤患者临床参数的相关性。结果:骨肉瘤组织hsa_circ_103809表达水平明显高于相应瘤旁组织,骨肉瘤细胞系hsa_circ_103809表达水平明显高于成骨细胞系hFOB1.19(均P<0.05)。转染后,实验组细胞hsa_circ_103809表达水平较对照组明显降低(P<0.01)。CCK-8实验结果显示,实验组细胞增殖能力较对照组明显减弱(P<0.01)。Transwell侵袭和迁移实验结果显示,实验组细胞侵袭和迁移能力均较对照组明显减弱(均P<0.01)。hsa_circ_103809表达水平与骨肉瘤患者临床分期及远处转移均呈正相关(均P<0.05)。结论:hsa_circ_103809高表达可促进骨肉瘤细胞增殖、侵袭和迁移。
Abstract:
Objective:To explore the expression and biological effect of circular RNA hsa_circ_103809 in osteosarcoma.Methods:qRT-PCR was used to detect the expression level of hsa_circ_103809 in osteosarocma tissues and cell lines.Osteosarcoma cells were divided into two groups.The experimental group was transfected with hsa_circ_103809 siRNA sequence.The control group was transfected with siRNA negative control(NC)sequence.CCK-8 and Transwell invasion and migration assays were performed to observe the abilities of proliferation,invasion and migration of the two groups.The correlation between hsa_circ_103809 expression level in osteosarcoma tissues and the clinical parameters of patients with osteosarcoma were analyzed.Results:Hsa_circ_103809 expression level in osteosarcoma tissues was higher than that in corresponding adjacent normal tissues,and the expression level of hsa_circ_103809 in osteosarcoma cell line was significantly higher than that in osteoblast cell line hFOB1.19(all P<0.05).After transfection,compared with the control group,the expression level of hsa_circ_103809 in the experimental group was significantly down-regulated,and the abilities of proliferation,invasion and migraiton were significantly weakened(all P<0.01).The expression level of hsa_circ_103809 was positively correlated with the clinical stage and distant metastasis of patients with osteosarcoma(all P<0.05).Conclusion:High expression of hsa_circ_103809 can promote the proliferation,invasion and migration of osteosarcoma cells.
