目的:探究长链非编码RNA SNHG15在鼻咽癌中的表达情况,并进一步分析其下游miRNA调控通路,为研究鼻咽癌的分子生物学行为及潜在治疗靶点提供参考。方法:采用PCR检测SNHG15在鼻咽癌临床组织标本及鼻咽癌细胞系中的表达水平。根据临床资料分析SNHG15表达与鼻咽癌患者预后的相关性。采用CCK-8实验、克隆形成实验以及流式细胞仪检测SNHG15对鼻咽癌细胞增殖能力与凋亡情况的影响。通过双荧光素酶实验、Western blot检测SNHG15、与SNHG15结合的miRNA以及潜在下游靶基因Krüppel样因子9(KLF9)之间的调控关系。结果:SNHG15在鼻咽癌组织和鼻咽癌细胞系中的表达上调,而SNHG15的相对表达量增高也预示着鼻咽癌患者的不良预后。在鼻咽癌细胞系中抑制SNHG15的表达能够显著抑制细胞的增殖能力,并促进鼻咽癌细胞的凋亡。双荧光素酶报告基因实验、qRT-PCR以及Western blot结果显示,在鼻咽癌细胞中SNHG15能够吸附并抑制miR-141-3p,并可能进一步调控转录因子KLF9的功能,从而促进鼻咽癌细胞的增殖。结论:在鼻咽癌中,SNHG15可能通过影响miR-141-3p/KLF9通路促进鼻咽癌细胞的增殖,参与疾病的发生与发展。
Abstract:
Objective:To investigate the expression of SNHG15 in nasopharyngeal carcinoma(NPC)and further analyze its downstream miRNA regulatory pathway,so as to provide reference for the study of molecular biological behavior and potential therapeutic targets of NPC.Methods:The relative expression level of SNHG15 was detected by real-time fluorescent quantitative PCR(qRT-PCR).The correlation between the expression level of SNHG15 and the prognosis of NPC patients was analyzed according to clinical data.The effects of SNHG15 on the proliferation and apoptosis of NPC cells were detected by CCK-8,clone formation test and flow cytometry.Through bio-informatics analysis,miRNA and potential downstream target genes that can bind to SNHG15 were found.The regulation relationship among them was detected by double luciferase and Western blot.Results:The expression of SNHG15 was significantly up-regulated in NPC tissues and cell lines,and the increased expression of SNHG15 indicated the poor prognosis of NPC patients.Inhibition of SNHG15 expression in NPC cell lines could significantly inhibit cell proliferation and promote apoptosis of NPC cells.The results of double luciferase reporter gene,qRT-PCR and Western blot showed that SNHG15 could competitively bind miR-141-3p with KLF9 in NPC cells,which may be the potential molecular mechanism of SNHG15's role in NPC.Conclusion:In NPC,SNHG15 may promote the proliferation of NPC cells by affecting miR-141-3p/KLF9 pathway,and participate in the occurrence and development of disease.
