摘要:
目的:探讨Purmorphamine对脑缺血再灌注大鼠细胞凋亡的影响。方法:采用线栓法制作大脑中动脉闭塞模型,将实验大鼠随机分为假手术组、模型组、PM低剂量组、PM中剂量组、PM高剂量组,于术后24 h, 采用TTC染色测脑梗死体积,TUNEL染色检测脑组织细胞凋亡,Western blot 测定B淋巴细胞瘤-2基因(Bcl-2)、Bcl-2相关X蛋白(Bax)的含量。结果:各药物组脑梗死体积明显低于模型组(P<0.05)。各药物组细胞凋亡数与模型组相比较明显降低(P<0.05),其中PM高剂量组降低最为显著。各药物组与模型组相比较,Bcl-2蛋白明显上调,凋亡基因Bax表达明显下调(P<0.05),其中PM高剂量组变化最为显著。结论:Purmorphamine在脑缺血再灌注中可以通过抗细胞凋亡从而减轻脑损伤。
Abstract:
Objective:To investigate the effect of Purmorphamine on apoptosis in rats with cerebral ischemia-reperfusion.Methods:The middle cerebral artery occlusion model was made by the suture method,and the experimental rats were randomly divided into sham operation group,model group,low-dose PM group,medium-dose PM group,and high-dose PM group.24 hours after surgery,TTC staining was used to measure cerebral infarction volume,TUNEL staining was used to detect cell apoptosis in brain tissues,and Western blot was used to measure the contents of Bcl-2 and Bax.Results:The cerebral infarction volume of each drug group was significantly lower than that of the model group(P<0.05).Compared with the model group,the number of apoptotic cells in each drug group was significantly reduced(P<0.05),and the decrease was most significant in the high-dose PM group.Compared with the model group,the Bcl-2 protein was significantly up-regulated and the expression of the Bax was significantly down-regulated(P<0.05),and the high-dose PM group had the most significant changes.Conclusion:Purmorphamine can reduce brain injury through anti-apoptosis during cerebral ischemia-reperfusion.
