摘 要 〖HT5K〗目的: 研究重楼皂苷 I (PPL I)对小鼠成骨样细胞MC3T3E1增殖的作用及其机制。方法: 采用MTT比色法、EdU试剂盒、碱性磷酸酶(ALP)活性检测法、茜素红染色法,观察加入PPL I (1、3、10、30 μmol/L)后,MC3T3E1的增殖、成熟与矿化情况;采用免疫印迹法,观察PPL I (3、10 μmol/L)对细胞中β连环素(βcatenin)和骨形成蛋白2 (BMP2)的表达影响。结果: PPL I可以有效地促进成骨样细胞MC3T3E1增殖,并浓度依赖性地升高βcatenin与BMP2蛋白的水平。 结论: PPL I能促进MC3T3E1细胞的增殖与分化,可能与其调节βcatenin与BMP2的表达有关。
Abstract:
Abstract 〖WT5BZ〗〖HT5”SS〗Objective:To investigate the effect of polyphyllin I on the proliferation of osteoblastlike cells MC3T3E1 and the possible mechanisms. Methods:The osteoblastlike cells MC3T3E1,were administered with different concentrations of polyphyllin I (1,3,10,and 30 μmol/L). Then,MTT colorimetric test method,EdU kit,alkaline phosphatase (ALP) activity detection kit,and Alizarin RedS stain were used to observe the proliferation and mature of MC3T3E1;Western blot analysis was utilized to observe the expression of βcatenin and bone morphogenetic protein 2 (BMP2) in MC3T3E1 cells treated with 3 and 10 μmol/L of polyphyllin I. Results:Polyphyllin I could effectively promote MC3T3E1 cells proliferation and upregulate the expression of βcatenin and BMP2 expression. Conclusion:Polyphyllin I may promote MC3T3E1 cell proliferation and differentiation via regulating βcatenin and BMP2 signaling pathway.
