摘要:
摘 要 目的:观察加巴喷丁对感染后咳嗽(PIC)大鼠肺泡灌洗液(BALF)和肺组织瞬时受体电位香草酸亚型1(TRPV1)、P 物质(SP)、降钙素基因相关肽(CGRP)含量的影响,探讨加巴喷丁对PIC气道神经源性炎症的干预作用。方法:60只大鼠随机分为三组:正常组、模型组和西药组(加巴喷丁组)。依据文献方法建立PIC大鼠模型。每组大鼠经过相应处理共10 d,观察3 min内大鼠咳嗽次数的变化,各组大鼠BALF、血清中TRPV1、SP、CGRP含量采用ELISA法检测,肺组织TRPV1、SP、CGRP蛋白表达采用免疫组化检测,并进行半定量评分,支气管和肺组织标本行HE染色,光镜观察其病理改变。结果:①咳嗽次数比较:模型组大鼠咳嗽次数明显增多,与正常组相比,差异有统计学意义(P<0.05);经西药干预后,大鼠咳嗽次数明显减少,与模型组比较,差异有统计学意义(P<0.05)。②各组大鼠BALF中TRPV1、SP、CGRP含量比较:正常组大鼠BALF中TRPV1、SP、CGRP均轻度表达,模型组以上各指标均明显升高,与正常组比较,差异有统计学意义(P<0.05); 经西药干预后,西药组大鼠BALF中TRPV1、SP、CGRP表达均减低,与模型组比较,差异有统计学意义(P<0.05)。各组大鼠肺组织病理学变化:正常组支气管黏膜无水肿,可见少量炎性细胞浸润。模型组支气管黏膜水肿,上皮脱落,肺泡壁轻度水肿,上皮变性、坏死、脱落,间质充血、水肿,可见大量炎性细胞浸润。西药组病理改变类似模型组,但总体炎性改变较模型组轻。各组大鼠肺组织TRPV1、SP、CGRP免疫组化检测结果比较:与正常组比较,模型组、西药组大鼠肺组织TRPV1、SP、CGRP蛋白均升高,差异有统计学意义(P<0.05);与模型组比较,西药组大鼠肺组织TRPV1、SP、CGRP蛋白表达减低,差异有统计学意义(P<0.05)。结论:加巴喷丁能够减少PIC大鼠BALF和肺组织TRPV1、SP、CGRP的表达,可能是通过干预TRPV1、SP、CGRP来降低PIC气道神经源性炎症。
Abstract:
Abstract Objective:To observe the effects of gabapentin on transient receptor potential vanilloid 1 (TRPV1),substance P (SP) and calcitonin gene-related peptide(CGRP) of Bronchoalveolar lavage fluid (BALF) and Lung tissue in post-infected cough (PIC) rats. To investigate the effect of gabapentin on neurogenic inflammation of PCI airway. Methods: Sixty rats were randomly divided into three groups: normal group,model group and western medicine group (gabapentin group). A PIC rat model was established according to literature methods. Each group of rats was treated for 10 days. The changes of cough frequency in rats within 3 min were observed. The contents of TRPV1,SP and CGRP in BALF and serum of each group were detected by ELISA. The expression of TRPV1,SP and CGRP protein in lung tissue was detected. Immunohistochemical examination was performed and semi-quantitative scoring was performed. HE staining was performed on HE staining of bronchial and lung tissue specimens,and pathological changes were observed by light microscopy. Results:①Comparison of cough times: The number of co〖LM〗ughs in the model group increased significantly. Compared with the normal group,the difference was statistically significant (P<0.05). After western medicine intervention,the number of coughs in the rats was significantly reduced,compared with the model group. The difference was statistically significant (P<0.05). ②Comparison of TRPV1,SP and CGRP contents in BALF of rats in each group: TRPV1,SP and CGRP in BALF of normal group were slightly expressed,and all indexes above the model group were significantly increased. Compared with the normal group,the difference was statistically significant. The significance of learning (P<0.05); after western medicine intervention,the expression of TRPV1,SP and CGRP in BALF of western medicine group were decreased,compared with the model group,the difference was statistically significant (P<0.05). ③Pathological changes of lung tissue in each group: bronchial mucosa in the normal group was edema,and a small amount of inflammatory cell infiltration was observed. In the model group,bronchial mucosa edema,epithelial shedding,mild alveolar wall,epithelial degeneration,necrosis,shedding,interstitial hyperemia,edema,and a large number of inflammatory cell infiltration. The pathological changes of the western medicine group were similar to the model group,but the overall inflammatory changes were lighter than the model group. ④Comparison of TRPV1,SP and CGRP immunohistochemical results in lung tissue of each group: Compared with the normal group,the TRPV1,SP and CGRP proteins in the lung tissue of the model group and the western medicine group were increased,and the difference was statistically significant ( P<0.05);Compared with the model group,the expression of TRPV1,SP and CGRP protein in the lung tissue of the western medicine group was decreased,and the difference was statistically significant (P<0.05). Conclusion: Gabapentin can reduce the expression of TRPV1,SP and CGRP in BALF and lung tissue of PIC rats. It may be through intervention of TRPV1,SP and CGRP to reduce neurogenic inflammation of PCI.
