摘 要 〖HT5K〗目的: 探讨IL-10在大鼠蛛网膜下腔出血(SAH)后早期脑损伤中的表达及作用。方法: 选择雄性SD大鼠60只,将其随机分为假手术组、SAH 1 d组、SAH 3 d组、SAH 1 d+IL-10组、SAH 3 d+IL-10组,每组各12只。SAH模型通过颈内动脉穿刺法建立,IL-10在SAH建模后30min内通过立体定向侧脑室穿刺给予。利用免疫组化染色和Western blot检测SAH后IL-10的表达,通过尼氏染色和HE染色评估IL-10对SAH早期脑损伤的作用,通过Western blotting检测TNF-α和IL-1β的变化,通过免疫组化染色评估SAH后小胶质细胞变化,并对大鼠进行行为学评分。结果:SAH 1 d组大鼠脑组织Wills环周围蛛网膜下腔大量出血,HE染色可见皮层组织疏松,神经元肿胀、核变形边集;SAH 1 d组及SAH 3 d组IL-10表达比较无统计学差异(P= 0.5669,P=0.9398)。与假手术组对比,SAH 1 d和SAH 3 d组TNF-α及IL-1β水平明显升高(P<0.0001)。SAH后小胶质细胞明显活化。IL-10可缓解SAH 1 d组神经元损伤,减少SAH 1 d组及SAH 3 d组TNF-α及IL-1β的表达(P<0.0001),显著减少小胶质细胞活化。结论:炎症反应在SAH早期脑损伤中起重要作用,SAH后小胶质细胞迅速激活,并通过释放TNF-α、IL-1β等炎症因子造成损伤。IL-10可通过缓解小胶质细胞活化,减少炎症因子的释放,改善SAH后早期脑损伤。
Abstract:
Abstract Objective: To explore the expression and role of IL-10 in early brain injury after subarachnoid hemorrhage (SAH) in rats. Methods:Sixty SD male rats were randomly divided into sham group, SAH 1 d group, SAH 3 d group, SAH 1 d+IL-10 group, and SAH 3 d+ IL-10 group, 12 rats for each group. The SAH Model was established by an endovascular perforation technique. IL-10 was injected stereotactically into lateral ventricle. The injury after SAH was assessed by eosin hematoxylin staining (HE staining). The expression of IL-10 was detected by western blot and immunohistochemistry. The effect of IL-10 on early brain injury was assessed by HE staining and Nissl staining. The expression of TNF-α and IL-1β was detected by western blot. Microglia was labelled by Iba-1 immunohistochemistry. Results :There was obvious subarachnoid hemorrhage around wills ring in SAH 1 d group. Compared with sham group, tissue looseness, neuronal swelling and nuclear anomalies were obviously seen in SAH 1 d group. The expression of IL-10 didn’t change significantly compared with SAH 1 d group and SAH 3 d group (P= 0.5669,P=0.9398). Compared with sham group, the level of TNFα and IL-1β in SAH 1d group and SAH 3 d group were significantly increased (P<0.0001). The microglia were also activated after SAH. Based on Nissl staining and HE staining, IL-10 alleviated injuries induced by SAH. IL-10 also decreased expression of TNF-α and IL-1β (P<0.0001) and reduced microglial activation in SAH 1 d group and SAH 3 d group compared with SAH group.Conclusion: The expression of IL-10 is unchanged after SAH. Microglia are activated after SAH and release TNFα and IL-1β. IL-10 alleviates the inflammatory response and early brain injury after SAH.
