摘要:
摘 要 目的:探讨MDSCs对肺癌小鼠CD8+T细胞及生存期的影响。方法:选取C57小鼠15只行皮下接种LLC,采用随机数字表法将肿瘤大小符合规格的小鼠分为对照组和观察组,对照组小鼠尾静脉注射100μg兔IgG作为对照抗体,观察组小鼠尾静脉注射100μg Gr1删除抗体以删除荷瘤小鼠中的MDSC。采用流式细胞术(FCM)检测荷瘤小鼠外周血中MDSC和CD8+T细胞的变化,同时记录荷瘤小鼠肿瘤生长情况和生存期。结果:注射Gr1前两组荷瘤小鼠外周血中MDSC和CD8+T细胞比例无统计学差异(P>0.05);注射Gr1第3天,观察组MDSC比例为(1.58±0.30),显著低于对照组(38.35±3.22),CD8+T细胞比例为(9.47±0.83),显著高于对照组(4.23±0.47);注射Gr1第10天,MDSC比例为(31.95±3.60),显著低于对照组(63.67±5.77),CD8+T细胞比例为(5.82±0.56),显著高于对照组(2.88±0.34),差异具有统计学意义(P<0.05)。在荷瘤小鼠肿瘤生长25d左右,随着抗体的代谢消除,两组肿瘤大小差异无统计学意义(P>0.05)。观察组小鼠生存率显著高于对照组,差异具有统计学意义(P<0.05)。结论:高水平MDSCs能够降低CD8+水平,对生存期有较大影响,随着时间的延长,注射MDSCs抗体无法降低肿瘤生长速度。
Abstract:
Abstract objective:To investigate the effect of MDSCs on CD8+T Cells and survival time of lung cancer mice.Methods:15 case C57 mice were subcutaneously inoculated with LLC,Mice with tumor size were randomly divided into two groups: control group and observation group.In control group, 100μg rabbit IgG was injected into tail vein as control antibody.Mice in observation group were injected with 100μg Gr1 to delete antibody to remove MDSC from tumorbearing mice.The changes of MDSC and CD8+T cells in peripheral blood of tumorbearing mice were detected by flow cytometry.And Tumor growth and survival time were recorded in tumorbearing mice.Results:Before injection of Gr1,there was no significant difference in the proportion of MDSC and CD8 T cells in peripheral blood between the two groups (P>0. 05).On the third day of injection of Gr1,The ratio of MDSC in the observation group was (1.58 ±0.30) ,which was significantly lower than that in the control group (38.35 ±3.22).The ratio of CD8 T cells was (9.47 ±0.83), which was significantly higher than that in the control group (4.23 ±0.47).On the 10th day of injection of Gr1,The MDSC ratio was (31.95 ±3.60), which was significantly lower than that in the control group (63.67 ±5.77).The ratio of CD8 T cells was (5.82 ±0.56), which was significantly higher than that in the control group (2.88 ±0.34), and the difference was statistically significant (P<0. 05).After 25 days of tumor growth, there was no significant difference in tumor size between the two groups with the elimination of antibody metabolism (P>0.05).The survival rate in the observation group was significantly higher than that in the control group (P<0.05).Conclusion: high level of MDSCs can decrease the level of CD8+ and have a great influence on the survival time. With the extension of time, injection of MDSCs antibody can not reduce the growth rate of tumor.
