摘要:
摘 要 目的:研究二甲双胍对Eca109食管癌细胞迁移能力的影响及其可能机制。方法:MTT实验观察在盐酸二甲双胍作用下Eca109食管癌细胞的增殖能力;划痕实验观察盐酸二甲双胍作用24、48 h时Eca109食管癌细胞的迁移能力;Western blotting观察细胞侵袭、迁移相关蛋白MMP2、MMP9及GLUT1、MMP14的表达。结果:盐酸二甲双胍可在一定程度上抑制Eca109食管癌细胞的迁移能力,并降低GLUT1、MMP14、MMP2、MMP9的表达(P<0.05)。结论:二甲双胍可以降低Eca109食管癌细胞的迁移能力,可能是通过抑制GLUT1、MMP14、MMP2、MMP9通路而实现的。
Abstract:
Abstract Objective: To study the effect and mechanism of metformin on the migration ability of Eca109 esophageal carcinoma cells .Methods: MTT assay was used to observe the proliferation ability of Eca109 esophageal cancer cells under the action of metformin hydrochloride .The migration ability of Eca109 esophageal cancer cells at 24 h and 48 h was observed by scratch assay .Western blotting observed the expression of cell migration related protein like MMP2, MMP9 and protein GLUT1,MMP14.Results: Metformin hydrochloride can inhibit the migration ability of Eca109 esophageal cancer cells to a certain extent (P< 0.05). Metformin hydrochloride also can reduce the expression of GLUT1, MMP14, MMP2 and MMP9(P<0.05).Conclusion: Metformin can reduce the migration ability of Eca109 esophageal cancer cells, possibly by inhibiting the migration ability of GLUT1,MMP14 ,MMP2 and MMP9 pathway to reduce Eca109 esophageal cancer cells.
