摘 要 目的:探讨尿中阿尔茨海默病(AD)相关神经丝蛋白(AD7c-NTP)含量检测在AD诊断中的意义。方法:采用酶联免疫吸附测定法检测遗忘型轻度认知障碍(aMCI)患者38例、轻度AD(CDR1分)36例、中重度AD(CDR2~3分)32例患者尿液中AD7c-NTP含量,并选择智能正常的受试者30例为正常对照组,分析不同病程AD患者尿AD7c-NTP含量的差异。结果:三组患者尿液AD7c-NTP含量分别为1.87±0.78(0.44~1.79)ng/ml、2.25±0.27(1.72~3.41)ng/ml、3.42±0.35(1.21~2.36)ng/ml,与正常对照组比较,具有统计学差异(P<0.05或P<0.01);与aMCI组相比,轻度AD和中重度AD组尿AD7c-NTP含量明显增加,均具有统计学差异(P<0.05或P<0.01);轻度AD组与中重度AD组相比较,中重度AD组患者尿AD7c-NTP含量更高,有统计学差异(P<0.01)。结论:不同病程AD患者尿液中AD7c-NTP含量均增高,其增高的程度与痴呆程度相关。尿液中AD7c-NTP含量检测可作为AD辅助诊断的生物学指标。
Abstract:
Abstract Objective: To explore the clinical meanings of the Alzheimer-associated neuronal thread protein (AD7c-NTP) level in urine for the diagnosis of AD . Method: There were 38 cases with amnesic mild cognitive impairment (aMCI),36 cases with mild Alzheimer's disease (CDR=1), 32 cases with the Moderate to severe Alzheimer's disease(CDR≥2) and 30 healthy subjects(control group).Enzymelinked immunosorbent assay (ELISA) was used to detect the expression changes of AD7c-NTP in the four groups. Results: The levels of urine AD7c-NTP in aMCI group 、mild AD group and moderate to severe AD group were respectively 1.87±0.78(0.44~1.79)ng/ml、2.25±0.27(1.72~3.41)ng/ml、3.42±0.35(1.21~2.36)ng/ml, which were significantly higher than those of control group(P<0.05,P<0.01);urine AD7c-NTP levels in these two AD groups were higher than those in aMCI group while the Moderate to severe AD group was higher than the mild AD group.There were significant differences among these different stages of AD patients(P<0.05,P<0.01).Conclusion: The urine AD7c-NTP level increased during different processes of AD, which is associated with the degree of dementia.Urine AD7c-NTP level might be used as a biological indicator of auxiliary diagnosis of patients with AD.
