摘要:
目的:通过免疫荧光染色和透射电镜观察的方法,探索豚鼠精囊内是否存在Cajal间质细胞(ICC)。方法:制作豚鼠精囊组织切片并进行免疫荧光染色,使用酪氨酸激酶受体(c-Kit)抗体标记ICC,并使用神经元特异性烯醇化酶 (NSE)抗体标记神经纤维,α-actin抗体标记平滑肌细胞,观察ICC与神经、平滑肌之间的形态学关系。制作豚鼠精囊超薄组织切片,在透射电镜下寻找符合ICC超微结构特征的细胞,并观察其与周围细胞之间的超微结构联系。结果:免疫荧光染色标记发现豚鼠精囊壁平滑肌细胞间及精囊腔黏膜皱褶间质内有大量符合ICC一般形态特征的c-Kit阳性细胞,ICC和神经纤维、平滑肌细胞密切接触。透射电镜下发现豚鼠精囊组织内存在和c-Kit阳性ICC形态、分布一致,符合典型ICC超微结构特点的细胞。这些精囊ICC与神经末梢以及周围的平滑肌细胞之间关系紧密。结论:豚鼠精囊壁平滑肌细胞间及精囊腔黏膜皱褶间质内存在大量的特殊间质细胞,这些细胞表达ICC特异性抗原c-Kit并符合ICC超微结构特征,与神经纤维和平滑肌细胞关系密切。
Abstract:
Objective:To explore whether interstitial cells of Cajal (ICC) were found in guinea pig seminal vesicle with immunofluorescence and transmission electron microscopy. Methods:The guinea pig seminal vesicle slices were made for immunohistochemistry and anti-c-Kit was used to indentify ICC. The guinea pig seminal vesicle slices were made and the ultrastructural features and the relationship between ICC and surrounding cells were observed by transmission electron microscopy. Results:Immunohistochemistry revealed many c-Kit positive cells, conforming to the general morphological features of ICC, between smooth muscle bundles and in the honeycomb-like folds of guinea pig seminal vesicle. Ultrastructurally, many special interstitial cells had been found in accordance with the morphology and distribution of c-Kit positive ICC and the majority ultrastructural criterions of ICC. These cells were closely related with smooth muscle cells and nerve fibers. Conclusion:A large number of special interstitial cells are found between smooth muscle bundles and in the honeycomb-like folds of guinea pig seminal vesicle.
